胃癌中CXCR4与VEGFR2的表达及其相关性研究

摘　　要：目的探讨血管内皮生长因子受体2(VEGFR2)与趋化因子受体4(CXCR4)在胃癌组织中的表达及其临床意义。方法选取2012年1月至2015年12月潍坊医学院附属医院行胃癌手术的术后标本蜡块35例、上海芯超组织芯片补充病例75例和距胃癌组织5 cm的正常胃组织75例,利用组织芯片技术,应用免疫组织化学染色的方法,检测CX-CR4、VEGFR2蛋白在胃癌组织中的表达情况。结果有无淋巴结转移及不同临床分期胃癌组织中CXCR4蛋白表达比较,差异均有统计学意义(P<0.05);而不同年龄、性别、病理分化程度及肿瘤浸润深度胃癌组织中CXCR4蛋白表达比较,差异均无统计学意义(P>0.05)。不同肿瘤浸润深度及临床分期、有无淋巴结转移胃癌组织中VEGFR2蛋白表达比较,差异均有统计学意义(P<0.05);而不同年龄、性别及病理分化程度胃癌组织中VEGFR2蛋白表达比较,差异均无统计学意义(P>0.05)。胃癌组织中CXCR4蛋白表达与VEGFR2蛋白表达强度呈正相关(r=0.475,P=0.000)。结论 CXCR4与VEG-FR2蛋白在胃癌组织中均呈高表达;CXCR4与VEGFR2蛋白在胃癌组织中的表达与胃癌有无淋巴结转移及临床分期均呈正相关;VEGFR2与肿瘤浸润深度呈正相关;CXCR4与VEGFR2在胃癌组织中的表达强度也呈正相关。Objective To investigate the expressions of VEGFR2 and CXCR4 in gastric carcinoma tissue and their clinical significance.Methods Thirty-five cases of paraffin-embedded specimen after gastric carcinoma operation,75 supplement cases of Shanghai Xinchao tissue chip and 75 cases of normal gastric tissue with 5 cm from normal gastric carcinoma tissue in the Affiliated Hospital of Weifang Medical University from January 2012 to December 2015 were selected.The tissue chip technique and immunohistochemical staining were used to detect the expression of VEGFR2 and CXCR4 in gastric carcinoma tissue.Results The expression of CXCR4 protein had statistical difference between gastric carcinoma tissues with and without lymph node metastasis and among different clinical stages(P<0.05),but which had no statistical difference between different sexes,among different ages,pathological differentiation degrees and tumor infiltration depths(P>0.05);the expression of VEGFR2 protein had statistical difference among different tumor infiltration depths,clinical stages,and between the gastric carcinoma tissues with and without lymph node metastasis(P<0.05);while which had no statistical difference between different sexes,and among different ages and different pathological differentiation degrees(P>0.05).The CXCR4 protein expression in gastric carcinoma tissues had a positive correlation with the VEGFR2 protein expression(r=0.475,P=0.000).Conclusion CXCR4 and VEGFR2 proteins are highly expressed in gastric cancer tissues;the expression of CXCR4 and VEGFR2 proteins in gastric carcinoma tissues is positively correlated with lymph node metastasis and clinical stage;VEGFR2 is positively correlated with the tumor infiltration depth;CXCR4 also has a positive correlation with the expression intensity of VEGFR2 in gastric carcinoma tissues.
