摘  要:本文以亚硝基胍为诱变剂对出发菌株进行诱变处理,通过蓝色凝胶平板初筛及发酵液排油圈直径和产量复筛,选育出高产且遗传稳定的突变菌株NTG-6,其生物表面活性剂产量达到40.83 g/L,比出发菌株提高50.84%。该突变菌株所产生物表面活性剂可将水的表面张力降至29.18 m N/m,其临界胶束浓度为15 mg/L,24 h乳化值为67.5%。该突变菌株所产生物表面活性剂对大肠杆菌、金黄葡萄球菌以及枯草芽孢杆菌均表现出良好的抑菌活性,且抑菌性能优于吐温80、SDS和莎梵婷。The mutagenesis of the starting strain was treated with nitroguanidine as a mutagenic agent.A mutant strain NTG-6 with high biosurfactant-producing and stable heredity was bred by means of primary screening with blue gel screening plate,secondary screening with oil spreading test and product yield as indicator.The biosurfactant yield of mutant strain reached to 40.83 g/L,which was 50.84%higher than that of the original strain.The surface tension of water could be reduced to 29.18 mN/m by using surfactant.The critical micelle concentration and the emulsification property of 24 h of the biosurfactant was 15 mg/L and 67.5%respectively.Compared with Tween 80,SDS and Surfactin,the biosurfactant produced by the mutant showed favorable bacteriostatic activity against Escherichia coli,Staphylococcus aureus and Bacillus subtilis.
