摘　　要：目的探讨肝细胞癌(hepatocellular carcinoma,HCC)患者肿瘤组织中长链非编码RNA(long non-coding RNA,Lnc RNA)LOC728290的表达及其临床意义。方法应用实时荧光定量PCR(real-time quantitative PCR,q RT-PCR)检测HCC患者手术切除的癌组织及其配对癌旁组织中Lnc RNA LOC728290的表达水平;统计分析其表达水平与患者临床病理特征之间的相关性,同时结合患者生存时间及预后随访情况,分析Lnc RNA LOC728290表达水平和无复发生存率之间的关系。结果 83%(54/65)HCC患者癌组织中Lnc RNA LOC728290的表达量较配对癌旁组织下调(P<0.05);Lnc RNA LOC728290的表达水平在不同AFP水平、有/无微脉管浸润的患者中,呈显著差异表达(P<0.05);生存分析表明Lnc RNA LOC728290表达水平低的HCC患者无复发生存率较其表达水平高的患者显著降低(P<0.05)。结论 Lnc RNA LOC728290在HCC患者肿瘤组织中呈显著低表达,其有望成为临床HCC早期诊断、转移及预后评估的潜在分子标志物和治疗靶标。Objective To determine the expression of long non-coding RNA(LncRNA)LOC728290 in tumor tissue of hepatocellular carcinoma(HCC)patients and explore its clinical significance.Methods The expression level of LncRNA LOC728290 in surgically resected tumor tissues and adjacent non-tumor tissues of HCC patients was detected by using quantitative real-time PCR,and its correlation with clinicopathological characteristics was analyzed.Meanwhile,the correlation between LncRNA LOC72829 expression and recurrence-free survival was analyzed based on the survival period and prognosis follow-up.Results The expression of LncRNA LOC728290 in tumor tissues of 83%(54/65)HCC patients was significantly decreased compared with that in adjacent non-tumor tissues(P<0.05).A significant difference was observed in LncRNA LOC728290 expression among patients with different levels of serum AFP and patients with or without microvascular invasion(P<0.05).Survival analysis displayed that the recurrence-free survival rate in patients of lower expression of LncRNA LOC72829 was significantly decreased compared with that of high expression of LncRNA LOC72829(P<0.05).Conclusions LncRNA LOC72829 is significantly lowly expressed in tumor tissue of HCC patients.It may provide a potential molecular marker and therapeutic target for early clinical diagnosis and evaluating the metastasis and prognosis in HCC.
