摘  要:目的:采取左肝外叶切除术建立C57BL/6老年小鼠术后认知功能障碍(POCD)模型,并分析手术创伤对老年小鼠行为学的影响。方法:选取清洁级15月龄C57BL/6老年小鼠80只,参照随机数字表法分为手术组、假手术组、麻醉处理组以及空白对照组。手术实施前5 d,老年小鼠每天进行3次Morris水迷宫训练,确保它们能够很好地找到固定放置的隐藏平台。第6天进行左肝外叶切除术。手术后1、3、7 d应用空间探索实验评价老年小鼠空间记忆能力,记录老年小鼠穿台次数以及目标象限停留时间百分比。结果:术前训练期间,各组老年小鼠的空间记忆能力持续提高(P<0.05),第5天趋向稳定。与空白对照组比较,麻醉处理组老年小鼠术后1、3、7 d的穿台次数和目标象限停留百分比无明显变化,假手术组术后1、3 d以及手术组术后各观察时间点都明显下降(P<0.05)。与假手术组比较,手术组上述指标各时间点下降更为明显(P<0.05)。与本组术后1 d比较,麻醉处理组以及空白对照组在手术后3 d以及7d的变化无显著性差异(P>0.05);假手术组在术后3 d降至最低,至7 d恢复正常;而手术组在术后3 d以及7 d仍明显降低(P<0.05)。结论:C57BL/6老年小鼠左肝外叶切除术后认知功能下降;采用这一方法能有效建立POCD动物模型。Objective:To establish postoperative cognitive dysfunction(POCD)model with C57BL/6 elderly mice by using extrahepatic left lobectomy,and analyze the effects of surgical trauma on mice ethology.Methods:Eighty C57BL/6 mice aged 15 months were selected and divided into operation group,sham operation group,anesthesia group and control group by reference to random digital table method.Five days before operation,mice received Morris water maze training 3 times each day to ensure that they were able to find the hidden platform.The extrahepatic left lobectomy was performed on the sixth day.The space exploration experiment was carried out 1,3 and 7 days after operation to evaluate the spatial memory capacity of mice,and the number of crossing stage and the percentage of time spent on the target quadrant were recorded.Results:The spatial memory ability of mice in each group continued to improve during the period of preoperative training(P<0.05),and tended to be stable on the fifth day.Compared with the control group,no significant difference was found in the number of crossing stage and the percentage of time spent on the target quadrant in anesthesia group,but the two indicators decreased significantly in sham operation group 1 and 3 days after operation and in operation group all the time point after operation(P<0.05),and much more obviously decreased in operation group compared with the sham operation group at all the time point(P<0.05).Compared with the first day after operation,there was no significant difference in anesthetic group and control group on the third and seventh day after operation(P<0.05).The indicators reached the lowest level in sham operation group on the third day and recovered gradually on the seventh day after operation,while in the operation group,the significant difference still existed on the third day and the seventh day compared to that on the first day after operation(P<0.05).Conclusiosn:The cognitive function of C57BL/6 aged mice declined after extrahepatic left lobectomy,such surgery may be
