摘 要:目的:探讨微弧氧化制备的多孔TiO_2涂层修饰的钛植体对骨髓间充质干细胞(BMSCs)粘附和成骨分化的影响。方法:采用微弧氧化技术在纯钛表面制备涂层,运用扫描电子显微镜(SEM)和X射线衍射仪(XRD)检测涂层表面形貌、晶相。与BMSCs共培养,采用免疫荧光染色检测粘附相关蛋白整合素β1表达,活死细胞染色和MTT法检测材料细胞毒性。碱性磷酸酶(ALP)和茜素红(ARS)染色以及RT-PCR检测涂层对BMSCs成骨分化的影响。结果:采用微弧氧化技术处理后的钛板(MAO组)涂层表面是微米级多孔TiO_2,并且涂层无明显细胞毒性,另外还促进了BMSCs中整合素β1表达。ALP和ARS染色及半定量结果显示,与未处理的钛板(Ti组)相比,MAO组BMSCs的ALP和ARS染色结果更深,显著高于Ti组(P<0.05)。RT-PCR结果表明,随着时间的延长,MAO组BMSCs ALP和Osterix表达水平逐渐升高。结论:纯钛表面通过微弧氧化制备的多孔TiO_2涂层促进了BMSCs粘附和成骨分化,并且没有明显细胞毒性。Objective:To investigate the effect of porous TiO 2 modified titanium implant prepared by micro-arc oxidation on the adhesion and osteogenic differentiation in bone marrow mesenchymal stem cells(BMSCs).Methods:Micro-arc oxidation technology was used to prepare the coating on the pure titanium surface.The surface morphology and crystal phase of the coating were detected by SEM and XRD.After co-culture of material and BMSCs,the expression of adhesion related protein integrinβ1 was detected by immunofluorescence staining,in addition,the cytotoxicity of the material was detected by living dead cell staining and MTT.Alkaline phosphatase(ALP)and alizarin red S(ARS)staining assay and RT-PCR were used to evaluate osteogenic differentiation of the BMSCs.Results:The coating of the MAO group treated by micro arc oxidation was microporous TiO 2,moreover,the coating not only had no obvious cytotoxicity,but also promoted the expression of integrinβ1 in BMSCs.The results showed that both ALP and ARS staining and quantitative of the MAO group was significantly higher than the Ti group which was the untreated group(P<0.05).The results of RT-PCR showed that the expression level of BMSCs ALP and OSX was significantly increased in MAO group over time.Conclusions:The porous TiO 2 coating prepared by micro-arc oxidation on pure titanium surface enhanced adhesion and osteogenic differentiation of BMSCs.
