摘 要:目的研究组蛋白去甲基化酶KDM3A在乳腺癌及正常乳腺组织中的表达差异以及其与临床病理特征的关系。方法用于免疫组化的乳腺癌组织及正常乳腺组织均来源于沈阳医学院附属中心医院,收集2013~2015年的乳腺癌石蜡标本,患者均为女性。采用免疫组化SP法检测100例乳腺癌、30例正常乳腺组织中KDM3A的表达情况。分析KDM3A的表达与患者年龄、雌激素受体(ER)、孕激素受体(PR)、肿瘤分期、瘤体大小以及淋巴结转移等临床病理特征的关系。结果 KDM3A在乳腺癌组织中的阳性率为89.0%(89/100),在正常乳腺组织中的阳性率为66.7%(20/30),KDM3A在乳腺癌中的表达高于正常乳腺组织,差异有统计学意义(P<0.05);KDM3A的表达与瘤体的大小以及ER表达有相关性。结论乳腺癌组织中KDM3A呈高表达,且与肿瘤瘤体的大小以及ER表达有相关性,提示KDM3A可能在ER阳性乳腺癌中发挥促进乳腺癌生长及增殖作用。Objective To study the difference of the expression of histone demethylation enzyme KDM3A in breast cancer tissue and normal breast tissue and its relationship with clinicopathological characteristics.Methods The breast cancer tissues and normal breast tissues used in immunohistochemistry were derived from the Central Hospital Affiliated to Shenyang Medical College.The paraffin specimens of breast cancer were collected from 2013 to 2015,all of which were women.The expression of KDM3A in 100 cases of breast cancers and 30 cases of normal breast tissues was detected by using immunohistochemical SP method.The relationship between the expression of KDM3A and the clinicopathological features of patients′age,estrogen receptor(ER),progesterone receptor(PR),tumor stage,tumor size and lymph node metastasis were analyzed.Results The positive rate of KDM3A in breast cancer tissues was 89.0%(89/100),and the positive rate of KDM3A in normal breast tissue was 66.7%(20/30).The expression of KDM3A in breast cancer was higher than that of normal breast tissue,with statistically significant difference(P<0.05).The expression of KDM3A was related to the size of the tumor and the expression of ER.Conclusion The high expression of KDM3A in breast cancer tissue is related to the size of tumor and the expression of ER,suggesting that KDM3A may play a role in promoting the growth and proliferation of breast cancer in ER-positive breast cancers.
