热休克蛋白65对视网膜毛细血管粥样硬化的影响

摘　　要：目的探讨热休克蛋白65(heat shock protein 65,HSP65)对视网膜毛细血管粥样硬化的影响。方法 30只成年Wistar大鼠随机分为实验组(n=15)和对照组(n=15),实验组给予高脂高糖饮食建立视网膜毛细血管粥样硬化模型,对照组正常喂养,8周后测定两组大鼠血清与视网膜组织中HSP65表达情况,电子显微镜下观察视网膜毛细血管粥样硬化情况与斑块情况。结果实验组的总胆固醇、甘油三酯、低密度脂蛋白胆固醇均高于对照组,血糖值也高于对照组,差异均有统计学意义(均为P<0.05)。实验组与对照组的血清中HSP65含量分别为(45.66±8.24)μmol·L^(-1)和(20.67±9.44)μmol·L^(-1),实验组高于对照组(P<0.05);实验组视网膜中的HSP65蛋白相对表达量为0.56±0.11,高于对照组的0.10±0.02,差异有统计学意义(P<0.05)。对照组视网膜毛细血管内皮细胞层和弹力板均完整且贴附紧密;实验组视网膜毛细血管内膜有程度不等的增厚,管腔呈偏心性狭窄,内膜中细胞外基质增多。实验组视网膜毛细血管的血管面积、斑块面积与斑块比例均高于对照组,差异均有统计学意义(均为P<0.05)。结论 HSP65在视网膜毛细血管粥样硬化大鼠的血清与视网膜组织中均呈高表达,可导致视网膜毛细血管的血管面积、斑块面积与斑块比例增加。Objective To investigate the effects of heat shock protein 65(HSP65)expression on retinal capillary atheromatous sclerosis.Methods Totally 30 Wistar rats were randomly divided into experimental group(n=15)and control group(n=15),and the experimental group was given high fat diet to construct a model of retinal capillary atheromatous sclerosis,while the control group received normal diet.After 8 weeks,the expression levels of HSP65 in the serum and retina tissue of rats were detected.The retinal capillary arteries were separated for the observation of the retinal capillary atherosclerosis and plate conditions were observed under an electron microscope.Results The total cholesterol,triglyceride and low-density lipoprotein cholesterol values as well as the blood glucose in the experimental group were higher than those of the control group,and the difference were all statistically significant(all P<0.05).The serum HSP65 level was(45.66±8.24)μmol·L-1 in the experimental group and(20.67±9.44)μmol·L-1 in the control group,respectively,and the experimental group was higher than the control group(P<0.05);the relative expression of HSP65 protein in the experimental group was higher than that in the control group,and the difference was statistically significant(P<0.05).The pathological section showed that the retinal capillary artery endothelium of the control group was intact,the elastic lamina was intact,and the monolayer was close to the inner elastic plate;while in the experimental group,the thickness of the retinal capillary artery was thickened to the lumen with eccentric stenosis,and the extracellular matrix increased in the intima.The vascular area,plaque area and plaque proportion in the experimental group were higher than those in the control group,and the differences were statistically significant(all P<0.05).Conclusion The expression of HSP65 in the serum and tissue of rats with retinal capillary atheromatous sclerosis was up-regulated,leading to the increase of the vascular area,plaque area,and plaque rat
