摘  要:目的探讨氧化苦参碱对结肠癌大鼠IL-2、IL-10和NF-κBp65表达的影响。方法将大鼠随机分为对照组、模型组、美沙拉嗪组和氧化苦参碱组,除对照组外,其余三组均构建结肠癌大鼠模型。健康对照组和模型组给予蒸馏水灌胃,美沙拉嗪组给予美沙拉嗪灌胃,氧化苦参碱组给予氧化苦参碱肌注,治疗2周。治疗结束后进行HE染色,并观察肿瘤体积变化。应用免疫组化法检测NF-κBp65表达,ELISA法检测IL-2和IL-10表达。结果美沙拉嗪组和氧化苦参碱组大鼠HE染色提示癌细胞结构不清晰,无明显细胞核,并呈片状坏死现象。与模型组相比,美沙拉嗪组和氧化苦参碱组大鼠肿瘤体积明显缩小,且后者肿瘤体积明显小于前者(P<0.05)。NF-κBp65表达定位于上皮细胞核巨噬细胞。与对照组比较,模型组NF-κBp65表达显著增加,与模型组比较,氧化苦参碱NF-κBp65表达显著下降,差异均有统计学意义(P<0.05)。与对照组比较,模型组IL-2水平升高,IL-10水平呈下降趋势,与模型组相比,氧化苦参碱组IL-2水平下降,IL-10水平上升,差异均有统计学意义(P<0.05)。结论氧化苦参碱注射液可应用于结肠癌的治疗,其作用机制可能是通过抑制促炎因子IL-2生成、促进抑炎因子IL-10分泌,以及阻断NF-κBp65的激活,从而发挥治疗作用。Objective To study the effect of oxymatrine on the expression of IL2,IL-10 and NF-κBp65 in colon cancer rats.Methods Rats were randomly divided into:control group,model group,mesalazine group and oxymatrine group.Except the control group,colon cancer rat model was constructed in the remaining three groups.The healthy control group and the model group were treated with distilled water.The mesalazine group was treated with mesalazine,and the oxymatrine group was injected with oxymatrine.The treatment last 2 weeks.After treatment,HE staining was performed and tumor volume changes were observed.The expression of NF-κBp65 was detected by immunohistochemistry,and the expression levels of IL-2 and IL-10 were detected by ELISA.Results HE staining in mesalazine group and Oxymatrine group showed that the structure of cancer cells was not clear,and there were no obvious nuclei,but with patchy necrosis.Compared with the model group,the tumor volume in the mesalazine group and the oxymatrine group was significantly smaller than that in the model group,and the volume of tumor in the latter group was significantly smaller than that in the former group(P<0.05).The expression of NF-κBp65 was localized in the epithelial nuclei and macrophages.Expression of NF-κBp65 in the model group was increased as compared with the control group.Compared with the model group,the expression of NF-κBp65 decreased in oxymatrine group(P<0.05).Compared with the control group,the level of IL-2 in the model group increased,and the level of IL-10 showed a downward trend.Compared with the model group,the level of IL-2 in the oxymatrine group decreased,and the level of IL-10 increased(P<0.05).Conclusion Oxymatrine injection could be used in the treatment of colon cancer.Its mechanism may be correlated with reducing the generation of pro-inflammatory cytokine IL-2,promoting the secretion of anti-inflammatory cytokine IL-10 secretion,and blocking the activation of NF-κBp65,and thus playing a role in treatment.
