摘  要:目的:研究安胃汤对慢性萎缩性胃炎(CAG)模型大鼠细胞凋亡因子表达的影响。方法:采用MNNG法建立CAG大鼠模型,将60只大鼠随机分为空白对照组(A组)、模型组(B组)、胃复春组(C组)、安胃汤低剂量组(D组)、安胃汤中剂量组(E组)、安胃汤高剂量组(F组)各10只,A、B两组给予3ml生理盐水灌胃,C组给予胃复春悬浊液按照0.078g/100(g·d)的剂量灌胃。D、E、F组分别给予0.5、1.0、2.0g/100(g·d)剂量的安胃汤灌胃,连续治疗4周。显微镜下观察胃黏膜组织形态的改变,使用QRT-PCR法和Western-blot法检测胃黏膜组织中Bax、Bcl-2、Fas、FasL mRNA及蛋白的表达,TUNEL法检测胃黏膜细胞凋亡指数。结果:镜下显示,B组胃黏膜明显萎缩,固有腺体萎缩消失,D、E、F组胃黏膜结构完整度呈递增趋势,固有腺体萎缩程度降低,数目增加,明显优于B组。与B组相比,C、D、E、F组的Bcl-2 mRNA相对表达量和蛋白表达量升高,Bax、Fas、FasL mRNA相对表达量和蛋白表达量降低(P<0.05);与C组相比,E、F组的Bcl-2mRNA相对表达量和蛋白表达量升高,Bax、Fas、FasL mRNA相对表达量降低(P<0.05)。与B组相比,C、D、E、F组凋亡指数均下降(P<0.05);与C组相比,D、E、F组凋亡指数均下降,且随着剂量的升高,下降程度更明显(P<0.05)。结论:安胃汤治疗CAG大鼠,能够通过调节Bax、Bcl-2、Fas、FasL等细胞凋亡因子水平,抑制胃黏膜细胞凋亡,改善胃黏膜萎缩,效果与安胃汤剂量正相关。Objective:To study the effect of Anwei decoction on the expression of apoptosis factor in rats with chronic atrophic gastritis.Methods:The CAG rat model was established by MNNG method.60 rats were randomly divided into A,B,C,D,E and F group for each 10 rats.Group A、B were given 3 ml normal saline,Group C were given 0.078 g/100(g·d)Weifuchun suspension.Group D,E and F were given 0.5、1.0、2.0 g/100(g·d)Anwei decoction,4 weeks of continuous treatment.The morphological changes of gastric mucosa were observed under microscope.The expressions of Bax,Bcl-2,Fas,FasL mRNA and protein in gastric mucosa were detected by QRT-PCR and Western-blot.The apoptotic index of gastric mucosa was detected by TUNEL.Results:Microscopic examination showed that the gastric mucosa in group B was significantly atrophy,the atrophy of the native glands was disappearing,and the structural integrity of gastric mucosa in D,E and F groups showed an increasing trend.The atrophy of glands in the glands decreased and the number increased,which was significantly better than group B.Compared with the B group,the relative expression and protein expression of Bcl-2 mRNA in the C,D,E and F groups increased,and the relative expression of Bax,Fas and FasL mRNA decreased and the protein expression was reduced(P<0.05.Compared with the C group,the relative expression and protein expression of Bcl-2 mRNA in the group of E and F increased,and the relative expression of Bax,Fas and FasL mRNA decreased(P<0.05);compared with the B group,the apoptotic index in C,D,E and F groups decreased(P<0.05).Compared with the C group,the apoptotic index in the D,E and F groups decreased,and the decrease was more obvious with the increase of the dose(P<0.05).Conclusion:The treatment of CAG can reduce the apoptosis of gastric mucosa and improve the atrophy of gastric mucosa by regulating the levels of apoptosis factors of Bax,Bcl-2,Fas,FasL,and the effect is positively related to the dosage of Anwei decoction.
