摘  要:目的:研究参葵汤对卵巢癌模型大鼠淋巴细胞、免疫球蛋白及相关炎症因子的影响,并且探讨其抗癌的作用机制。方法:将卵巢癌细胞NUTU-19接种于腋窝皮下,制备卵巢癌大鼠模型,取90只模型大鼠随机分为模型组、顺铂组和中药组,每组30只;各治疗组均每两天腹腔注射给药1次,共5次。检测血中T淋巴细胞亚群(CD_4^+、CD_8^+)百分率并计算CD_4^+/CD_8^+;酶联免疫(ELISA)法测定血清中IgG、IgM,白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量。结果:经过参葵汤干预后,中药组血清中CD_4^+/CD_8^+与模型组和顺铂组相比显著增高,差异具有统计学意义(P<0.01,P<0.05);CD_4^+与模型组相比显著升高,差异具有统计学意义(P<0.01),但与顺铂组相比并无统计学意义(P>0.05);CD_8^+与模型组和顺铂组相比显著增高,差异具有统计学意义(P<0.01,P<0.05)。IgG、IgM水平与模型组和顺铂组相比显著增高,差异具有统计学意义(P<0.01,P<0.05)。白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量与模型组相比显著降低,差异具有统计学意义(P<0.01),但与顺铂组相比并无统计学意义(P>0.05)。结论:参葵汤能够提高卵巢癌模型大鼠免疫功能的作用,其机制可能与参葵汤能够显著降低模型大鼠体内相关炎症因子及提高CD_4^+/CD_8^+有关。Objective:To study the effect of Shenkui decoction on lymphocyte,immunoglobulin and related inflammatory factors in ovarian cancer model rats and to explore its anticancer mechanism.Methods:The ovarian cancer cell NUTU-19 was inoculated subcutaneously in the axillary fossa to prepare an ovarian cancer rats model.90 rats were randomly divided into model group,cisplatin group,and Chinese medicine group,30 rats in each group.Each treatment group was given intraperitoneal injection every two days,a total of 5 times.The percentage of T lymphocyte subsets(CD 4+,CD 8+)in blood was measured and CD 4+/CD 8+was calculated.IgG,IgM,interleukin-6(IL-6)and tumor necrosis factor-alpha(TNF-α)were measured by enzyme-linked immunosorbent assay(ELISA).Result:After intervention by Shenkui decoction,the serum levels of CD 4+/CD 8+in the traditional Chinese medicine group were significantly higher than those in the model group and the cisplatin group,the difference was statistically significant(P<0.01,P<0.05).CD 4+was significantly higher than the model group,the difference was statistically significant(P<0.01),however,there was no significant difference compared with the cisplatin group(P>0.05).CD 8+increased significantly compared with the model group and the cisplatin group,the difference was statistically significant(P<0.01,P<0.05).The level of IgG,IgM increased significantly compared with the model group and the cisplatin group,the difference was statistically significant(P<0.01,P<0.05).The level of IL-6 and TNF-αwere significantly lower than those in the model group,the difference was statistically significant(P<0.01).However,there was not statistically significant compared with the cisplatin group(P>0.05).Conclusion:Shenkui decoction can increase the immune function of ovarian cancer model rats,and its mechanism may be related to Shenkui decoction can significantly reduce the related inflammatory factors in the rat model and increase CD 4+/CD 8+.
