摘 要:目的检测结直肠癌组织中WWOX(WW domain containing oxidoreductase)基因的杂合性缺失,并探讨其与结直肠癌临床病理参数之间的关系。方法用PCR-聚丙烯酰胺凝胶电泳-银染法检测结直肠癌组织中WWOX基因3个微卫星位点D16S3029,D16S504,D16S3096的杂合性缺失。结果 24例结直肠癌组织中有8例(33.3%)存在一个或一个以上位点的杂合性缺失(LOH)。按浸润深度分组,浆膜内LOH阳性率为9.09%(1/11),浆膜及浆膜外LOH阳性率为53.85%(7/13),两者间差异有统计学意义(P<0.05)。结论 WWOX参与调控结直肠癌的发生发展,LOH可能是WWOX在肿瘤中失活的重要机制。Objective To detect the heterozygosity loss of WWOX(WW domain containing oxidoreductase)gene in colorectal cancer tissue,and to discuss its relationship with clinicopathological parameters of colorectal cancer.Methods Using PCR-polyacrylamide gel electrophoresis-silver dye method to detect the heterozygosity loss of 3 microsatellite sites of WWOX gene in colorectal cancer tissue,D16S3029,D16S504 and D16S3096.Results Among the 24 cases of colorectal cancer,8(33.3%)had heterozygous deletion(LOH)on one or more sites.Grouped by the infiltration depth,the LOH positive rate inside the serous membrane was 9.09%(1/11),and the positive rate on the serous membrane and outside the serous membrane was 53.85%(7/13),and the difference was statistically significant(P<0.05).Conclusion WWOX is involved in regulating the development of colorectal cancer,and LOH may be an important mechanism of WWOX inactivation in tumor.
