摘 要:[目的]脂氧合酶(LOX)是导致谷物陈化变质的关键因素,为了深入研究LOX对谷子陈化的影响,需对谷子中脂氧合酶(LOX)最适活性条件及影响因素进行测定。[方法]本研究以晋谷21号为主要试验材料,亚油酸作反应底物,采用紫外分光光度法测定谷子中脂氧合酶(LOX)活性的最适反应条件及影响因素。[结果]作物谷子中脂氧合酶(LOX)活性的最适pH为7,最佳反应温度为30℃,最适底物添加量为200μL,粗酶最适提取时间为2h,最佳粗酶液加入量/底物加入量为1∶6。[结论]此研究结果可以为今后对谷子脂氧合酶(LOX)的研究提供一定的技术借鉴与指导。[Objective]Lipoxygenase(LOX)plays a key role on accelerated aging of cereals.The optimal reaction conditions and related impact factors of lipoxygenase(LOX)on foxtail millet grains were determined to support further mechanism study of the aging effect of LOX on foxtail millet.[Methods]The optimal reaction condition and several impact factors of lipoxygenase(LOX)were investigated using UV spectrophotometers.Jingu 21 was selected as experimental grain material and linoleic acid was used as the substrate.[Results]The idea reaction condition of lipoxygenase(LOX)was determined as pH value at 7,temperature at 30℃,reaction substrate additive amount at 200μL,the extraction time of crude enzyme for 2 h,and the extraction additive amount/substrate amount ratio at 1∶6.[Conclusion]The results of present study provided technical support and guidance for further research oflipoxygenase(LOX)in foxtail millet.
