胰泰复方对胰腺纤维化大鼠腺泡细胞凋亡的影响

摘　　要：目的观察胰泰复方对胰腺纤维化大鼠腺泡细胞凋亡的影响。方法将60只SD雄性大鼠按随机数字表法分为空白组、模型组、清胰利胆颗粒组和胰泰复方高、中、低剂量组。腹腔注射13.3%L-精氨酸造模后各组给药,胰泰复方高、中、低剂量组分别给予胰泰复方22.32、11.16、5.58 g/kg,清胰利胆颗粒组给予清胰利胆颗粒2.678 g/kg,空白组、模型组均给予2 m L生理盐水。各组连续治疗30 d后取胰腺,用于TUNEL细胞凋亡检测及Caspase-3的Western blot和免疫组化检测。结果 TUNEL细胞凋亡检测结果显示,胰泰复方各治疗组凋亡细胞面密度明显少于模型组(P<0.05),胰泰复方中剂量组明显少于胰泰复方低剂量组(P<0.05)。Western blot和免疫组化检测Caspase-3蛋白表达结果显示,胰泰复方高、中剂量组与模型组比较下降显著(P<0.05),胰泰复方中剂量组比胰泰复方高剂量组下降亦显著(P<0.05)。结论减少胰腺腺泡细胞凋亡可能是胰泰复方延缓胰腺纤维化的机制之一。Objective To observe the effect of Yitai Compound on apoptosis of acinous cells in the rats with pancreatic fibrosis.Methods Sixty SD male rats were randomly divided into blank group,model group,Qingyi Lidan Granules group,and high,medium,low dose of Yitai Compound groups by random number table method.After creation of models by intraperitoneal injection of 13.3%L-arginine,all rats were given the treatment.The rats in high,medium,low dose of Yitai Compound groups were respectively given Yitai Compound 22.32,11.16,5.58 g/kg,Qingyi Lidan Granules group was given Qingyi Lidan Granules 2.678 g/kg,both blank group and model group were given 2 mL normal saline.The treatment lasted for 30 days.After treatment,the pancreas was taken for detecting the apoptosis by TUNEL and the expression of Caspase-3 by Western blot and immunohistochemmistry.Results The results of cell apoptosis by TUNEL showed that the area density of apoptosis in all Yitai Compound groups was significantly less than that of model group(P<0.05),which of medium dose of Yitai Compound group was less than that of low dose of Yitai Compound group(P<0.05).The expression of Caspase-3 by Western blot and immunohistochemmistry showed that high,medium dose of Yitai Compound groups were lower than that of model group(P<0.05),and medium dose of Yitai Compound group was lower than that of high dose of Yitai Compound group(P<0.05).Conclusion Reducing the apoptosis of acinous cells in pancreas may be one of the mechanisms for Yitai Compound delaying pancreatic fibrosis.
