miRNA 452-5P及miRNA 215-5P在结直肠癌中的表达及意义

摘　　要：目的研究miRNA 452-5P及miRNA 215-5P在结直肠癌中的表达及其与临床病理特征的关系,并分析其诊断价值。方法收集30组结直肠癌患者的结直肠癌组织及癌旁正常肠黏膜组织,采用实时荧光定量PCR技术检测全部患者结直肠癌组织及癌旁正常肠黏膜组织中miRNA 452-5p、miRNA 215-5p的表达,分析其与临床病理资料之间的关系,并进行ROC分析。结果 miRNA 452-5P在结肠癌组织中表达显著下调(P<0.001),miRNA 215-5p在结肠癌组织中表达显著上调(P<0.001),两种miRNA与结直肠癌患者年龄、性别、分化程度、肿瘤位置、浸润深度、淋巴转移、Dukes分期等均不相关。miRNA 452-5p曲线下面积AUC=0.859(95%CI:0.719～0.999),miRNA 215-5p曲线下面积为AUC=0.879(95%CI:0.757～1.0)。2-miRNAs panel曲线下面积为AUC=0.930(95%CI:0.833～1.0)。结论 miRNA 452-5p在结肠癌组织中表达显著下调,miRNA 215-5p在结直肠癌组织中表达显著上调。2-miRNAs panel对结直肠癌诊断价值较高,可作为潜在的结直肠癌诊断价值的分子标志物和治疗靶点。Objective To study the expression of miRNA 452-5P and miRNA 215-5P in colorectal cancer and the relationship with clinic-pathological characteristics.To analyze their diagnostic values.Methods A total of 30 groups of colorectal cancer tissues and normal intestinal mucosa tissues beside the cancer of colorectal cancer patients were collected.The expression of miRNA 452-5p and miRNA 215-5p in colorectal cancer tissues and nor-mal intestinal mucosa tissues beside the cancer of all patients was measured by real-time fluorescence quantitative PCR,and their relationship with clinic pathological data was analyzed and ROC analysis was implemented.Results The expression of miRNA 452-5P in colon cancer tissue was significantly decreased compared with that in normal control(P<0.001).The expression of miRNA 215-5p in colon cancer tissues was significantly in-creased compared with that in normal control(P<0.001).Two kinds of miRNA were not related to age,sex,degree of differentiation,tumor location,infiltration depth,lymphatic metastasis and Dukes staging of colorectal cancer patients.The area below the miRNA 452-5p curve is AUC=0.859(95%CI:0.719~0.999).The area un-der the curve(AUC)of the miRNA 215-5p is 0.879(95%CI:0.757~1.0).The area under the curve(AUC)of the 2-miRNAs panel curve is 0.930(95%CI:0.833~1.0).Conclusions The expression of miRNA 452-5p is sig-nificantly decreased in colon cancer tissues,and the expression of miRNA 215-5p is significantly increased in colorectal cancer tissues.2-miRNAs panel is of high diagnostic value for colorectal cancer and can be used as a potential molecular marker and therapeutic target for the diagnosis of colorectal cancer.
