摘 要:为获得具有木质素降解功能的放线菌菌株并研究其产酶特性,通过苯胺蓝平板和木质素平板从造纸厂污泥中分离得到一株木质素降解放线菌AG-11,其对木质素的降解率可达60%以上,经过16SrDNA测序鉴定为灰略红链霉菌(Streptomyces griseorubens),Genbank登陆号为KX774674。研究了AG-11在液体条件下的产酶特性,发现AG-11对碱性木质素的降解主要依靠胞外漆酶(Lac)和胞外木质素过氧化物酶(LiP),且产酶活性受到木质素降解中间产物的诱导,同时对金属离子表现出了较强的敏感性,氧化胁迫也能显著提高AG-11的产酶活力,在20mmol/L H_2O_2胁迫下,其胞外Lac及胞外LiP酶活可分别达25.42、26.81U/mL。In order to research the enzyme-production characteristics of lignin-degrading antinomyces,a lignin-degradation actinomycetes named as AG-11 was isolated from active-sludge by aniline blue plate and lignin plate method,its lignin degradation rate could reach to over 60%.The strain AG-11 was identified as Streptomyces griseorubens by 16S rDNA,and the Genbank accession number was KX774674.The enzyme-production characteristics showed that degradation of lignin mainly depended on extracellular laccase and lignin peroxidase from AG-11.Then enzyme-production ability was regulated by molecules inducer and was sensitive to metal ions.Strain AG-11 could significantly improve enzyme-producing ability under the oxidative stress,under 20 mmol/L of hydrogen peroxide,the extracellular laccase activity and lignin peroxidase activity were 25.42,26.81 U/mL respectively.
