摘 要:目的:检测细胞命运决定因子(DACH1)在血管性痴呆海马组织中的表达及气味训练和碱性成纤维细胞生长因子(bFGF)的干预作用,探讨不同干预方法对血管性痴呆大鼠神经再生及脑组织DACH1蛋白表达的影响。方法:雄性Wistar大鼠随机分为假手术组、模型组、训练组、bFGF组。采用2血管阻断加硝普钠降压法制作大鼠血管性痴呆(VD)动物模型,用H-E、TTC染色法和免疫组织化学方法检测海马组织nestin和DACH1蛋白的表达及气味训练和bFGF的干预作用。结果:脑缺血再灌注第7天缺血海马神经元nestin阳性细胞明显增多达高峰。DACH1在脑缺血再灌注第7天较正常组增多,随缺血再灌注时间的进一步延长逐渐增多,第21天持续高水平表达。气味训练和bFGF组nestin和DACH1蛋白的表达明显增加。结论:气味训练和bFGF促进神经干细胞的增殖分化以及缺血脑组织DACH1蛋白的表达,提示气味训练和bFGF促进神经干细胞增殖分化作用可能由DACH1信号通路介导。Objective:To investigate the expression of cellfate determination factor dachshund1(DACH 1)and hippocampal neurogenesis in a rat model of cerebral ischemia/reperfusion,and to explore bFGF and odor training mechanism.Methods:Wistar rats were randomized into sham groups ischemia and reperfusion group,training group and bFGF group.Odor training was performed at day 1 after the ischemia and reperfusion.The model of two vessel occlusion plus sodium nitroprusside was established.The expression of nestin and DACH1 in the hippocampus was detected with TTC,H-E and immunohistochemical methods.Results:After 3 days,nestin positive cells in the hippocampus were obviously increased.On day 7,nestin positive cells were more than those at any time.After 7 days,DACH1 positive cells in the hippocampus were increased.DACH1 reaction products were increased gradually with the time of cerebral ischemia/reperfusion,reached the peak on 21 day.After odor training and bFGF,the expression of hippocampus nestin and DACH1 had apparently increased in each group.Conclusion:Odor training and bFGF promote the expression of nestin and DACH1 in the hippocampus tissues.These findings indicate that odor training and bFGF promotion of neural stem cell proliferation and differentiation may be mediated by DACH1 signaling pathway.
