MiRNA-101在结直肠腺瘤及结直肠癌中表达差异的分析

摘　　要：目的检测miRNA-101在结直肠腺瘤及结直肠癌组织中的表达差异并探讨其临床意义。方法收集33例结直肠腺瘤组织(管状绒毛腺瘤8例,管状腺瘤25例)及瘤旁正常组织,31例结直肠癌组织及癌旁正常组织,采用荧光定量PCR技术测定miRNA-101的相对表达量,分析其与年龄、性别、病理类型等之间的关系。结果miRNA-101在腺瘤中的表达高于瘤旁组织(P<0.05),在癌组织中的表达低于癌旁组织(P<0.05),在不同年龄、性别、病理类型及肿瘤部位的相对表达量均无差异,无统计学意义。结论miRNA-101在腺瘤组织中表达升高,在结直肠癌组织中表达降低,提示其在正常结直肠组织到结直肠腺瘤再到结直肠癌的发生发展过程中发挥了重要的调控作用。Objective:To investigate the expression of miRNA-101 in colorectal cancer and colorectal adenomas polyp.Methods:33 colorectal adenomas polyp specimens and paired parapolyp tissues(including 8 tubulovillous adenoma and 25 tubular adenoma)and 31 colorectal cancer specimens and paired paracancer tissues were collected for assessment of miR-101 by real-time quantitative PCR,analyzed the relationship between the expression of age,gender,pathological type and other clinical significance.Results:The expression of miRNA-101 in colorectal adenomas polyp tissues is higher than in paired parapolyp tissues(P<0.05),and lower in colorectal cancer tissues than in paired paracancer tissues(P<0.05);But no significant difference among age,gender,pathological types(tubulovillous adenoma and tubular adenoma)and tumor locations.Conclusions:The different expression level of miRNA-101 in colorectal adenomas polyp and colorectal cancer has been linked to colorectal.
