青蒿素对肾癌细胞株786-0细胞的增殖和Fascin表达的影响

摘　　要：目的:探讨青蒿素对肾癌细胞株786-0细胞的增殖和Fascin表达的影响。方法:选用786-0肾癌细胞株,分别通过不同浓度的青蒿素作用细胞株,收集不同时间点(0、24、48、72 h)的细胞,运用细胞增殖CCK-8法测定青蒿素对肾癌细胞株786-0细胞的增殖;通过Western Blot检测青蒿素干预后肾癌细胞株786-0细胞Fascin的表达。结果:随着药物浓度的增加,青蒿素可明显抑制肾癌细胞株786-0细胞的增殖(P<0.01),青蒿素可显著降低肾癌细胞株786-0 Fascin蛋白的表达(P<0.05)。结论:青蒿素通过抑制肾癌细胞增殖和Fascin蛋白的表达,从而抑制肿瘤细胞的恶性增生。To investigate the effect of Artemisinin on the proliferation and Fascin expression of human renal cell carcinoma cell line786-0.Method:786-0cell line were cultured in vitro and treated with different doses Artemisinin.The cells were collected at different time points(0,24,48,72h).CCK-8method was used to detect the cell proliferation,the expression of Fascin in renal cell carcinoma cell line786-0cells were analyzed by Western Blot.Result:Different doses artemisinin could significantly inhibit the proliferation of renal cell carcinoma cell line786-0with the increase of drug concentration(P<0.01),Artemisinin could obviously reduced the expression of Fascin in renal cell carcinoma cell line786-0(P<0.05).Conclusion:Artemisinin might inhibit the proliferation and the expression of Fascin,then inhibit the malignant proliferation of tumor cells
