摘 要:目的 探讨miRNA-149在膀胱癌组织中的表达及其生物意义。方法 采用qRT-PCR法检测miRNA-149在临床膀胱癌配对组织及膀胱癌细胞中的表达情况;采用PolyJet?DNA In Vitro Transfection Reagent将miRNA-149过表达质粒稳定转染膀胱癌细胞T24、T24T,并用荧光定量PCR技术检测转染率;利用Transwell法检测细胞的迁移和侵袭能力。结果 miRNA-149在侵袭性T24、T24T细胞中较膀胱上皮永生化细胞明显降低;同时在14例配对患者中,与癌旁正常组织相比,在膀胱癌组织中miRNA-149表达明显降低,与细胞水平呈一致趋势,差异有统计学意义(P<0.05);miRNA-149稳定过表达的T24、T24T细胞中发现其可显著抑制膀胱癌细胞侵袭而对膀胱癌细胞迁移无明显抑制作用,差异有统计学意义(P<0.05)。结论 miRNA-149在膀胱癌组织和细胞中异常表达,且miRNA-149表达上调可能抑制膀胱癌细胞侵袭,为膀胱癌的转移研究提供了新的生物学意义。Objective To investigate the expression of miRNA-149 in bladder cancer and its biological significance.Methods The expression of miRNA-149 in paired tissues and bladder cancer cells was detected by qRT-PCR.The miRNA-149 over-expression plasmid transfected bladder cancer cells T24,T24T by PolyJet?DNA In Vitro Transfection Reagent,and the transfection rate was detected by fluorescence quantitative PCR.The migration and invasiveness of cells were detected by Transwell method.Results The expression of miRNA-149 in invasive T24 and T24T cells were significantly lower than that in bladder epithelial immortalized cells;in 14 matched patients,the expression of miRNA-149 in bladder cancer tissue was significantly lower than that in adjacent normal tissue,with statistically significant differences(P<0.05).In miRNA-149 stably over-expressed T24 and T24T cells,miRNA-149 could significantly inhibit the invasion of bladder cancer cells,but could not significantly inhibit the migration of bladder cancer cells,with statistically significant differences(P<0.05).Conclusions miRNA-149 is abnormally expressed in bladder cancer tissue and cells,and the up regulation of miRNA-149 may inhibit invasion of bladder cancer cells,which provides a new biological significance for the study on bladder cancer metastasis.
