摘　　要：目的探究C595对人肺癌A549细胞株抑制增殖和诱导凋亡作用。方法 MUC1蛋白核心的重复序列IgG3单克隆抗体(C595)经免疫荧光标记及流式细胞术检测,分析C595对人肺癌A549细胞株抑制增殖和诱导凋亡。结果 A549细胞膜表面覆盖97.3%MUC1表达量,阴性对照组覆盖MUC1表达量1.03%;C595浓度增加,A549细胞株增殖抑制效果越强,凋亡率增加,存活率降低。结论单克隆抗体C595在体外抑制人肺癌细胞A549生长,C595浓度增加,A549细胞存活率降低,凋亡率增加。Objective To investigate the effect of C595 on proliferation and apoptosis of human lung cancer A549 cell line.Methods Immunofluorescent labeling and flow cytometry were performed on the IgG3 monoclonal antibody(C595)of the MUC1 protein core sequence.The effect of C595 on proliferation and induction of apoptosis in human lung cancer A549 cell lines was analyzed.Results The surface of A549 cell membrane covered 97.3%of MUC1 expression.The negative control group covered MUC1 1.03%;C595 concentration increased,the stronger the inhibition of A549 cell proliferation,the higher the apoptosis rate,and the lower the survival rate.Conclusion The monoclonal antibody C595 inhibits the growth of human lung cancer A549 in vitro.If the concentration of C595 increases,the survival rate of A549 cells decreases and the apoptosis rate increases.
