宫颈癌组织中卵泡抑素样蛋白1的表达变化及其意义

摘　　要：目的观察卵泡抑素样蛋白1(FSTL1)在宫颈癌组织中的表达变化,并探讨其意义。方法分别采用实时荧光定量PCR技术和免疫酶标法检测宫颈癌组织、宫颈癌旁组织及正常宫颈组织中的FSTL1 mRNA和蛋白。结果宫颈癌组织、宫颈癌旁组织、正常宫颈组织中FSTL1 mRNA的相对表达量分别为0.36±0.05、2.15±0.40、2.52±0.50,FSTL1蛋白的相对表达量分别为(3.33±0.24)、(4.76±0.25)、(5.41±0.22)分,宫颈癌组织中FSTL1mRNA和蛋白的表达量与宫颈癌旁组织及正常宫颈组织中的表达量相比,P均<0.05。宫颈癌组织中FSTL1 mRNA的表达与肿瘤大小、浸润深度、分化程度以及FIGO分期有关(P均<0.05),FSTL1蛋白的表达与肿瘤分化程度、浸润深度有关(P均<0.05)。宫颈癌组织中FSTL1蛋白高表达和低表达患者的总体生存时间(分别为20、26个月)和无瘤生存时间(分别为21、27个月)相比,P均>0.05。结论宫颈癌组织中FSTL1 mRNA和蛋白均呈低表达,观察癌组织中FSTL1 mRNA和蛋白的表达有助于判断宫颈癌的浸润深度、分化程度、肿瘤大小及FIGO分期,FSTL1可能作为抑癌基因参与宫颈癌的发生发展。Objective To explore the expression of follistatin-like protein 1(FSTL1)and to discuss its significance in cervical cancer tissues.Methods The expression of FSTL1 mRNA and protein in the cervical cancer tissues,adjacent tissues,and normal cervical tissues was detected by RT-PCR and immunohistochemistry,respectively.Results The relative expression levels of FSTL1 mRNA in the cervical cancer tissues,adjacent tissues,and normal cervical tissues were 0.36±0.05,2.15±0.40,and 2.52±0.50,respectively.The expression levels of FSTL1 protein in the cervical cancer tissues,adjacent tissues,and normal cervical tissues were 3.33±0.24,4.76±0.25,and 5.41±0.22,respectively.The mRNA and protein expression levels of FSTL1 in the cervical cancer was less than that of the adjacent tissues and normal cervical tissues(both P<0.05).The expression level of FSTL1 mRNA was associated with tumor maximum diameter,infiltration depth,degree of differentiation,and FIGO stage(all P<0.05),and FSTL1 protein was associated with tumor differentiation and tumor size(both P<0.05).Log-rank test showed that the overall survival time in the patients with FSTL1 protein high and low expression was 20 months and 26 months,respectively;the tumor-free survival time was 21 months and 27 months,respectively;there was no statistically significant difference between them(both P>0.05).Conclusion The expression of FSTL1 mRNA and protein is significantly down-regulated in the cervical cancer tissues.The expression of FSTL1 mRNA and protein in the cancer tissues is helpful to determine the infiltration depth,differentiation degree,tumor size,and FIGO stage of cervical cancer.FSTL1 may be a tumor suppressor gene and be involved in the occurrence and development of cervical cancer.
