摘  要:目的探讨大肿瘤抑制因子2(LATS2)基因启动子甲基化在口腔鳞状细胞癌(口腔鳞癌)发生、发展中的作用。方法选取108例口腔鳞癌患者手术切除的肿瘤组织,其中45例留取癌旁正常组织。采用RT-PCR法检测口腔鳞癌组织及癌旁正常组织LATS2 mRNA表达;采用特异性PCR法检测口腔鳞癌组织LATS2基因启动子甲基化情况,采用Spearman相关分析法分析口腔鳞癌组织LATS2启动子甲基化与患者临床病理参数的关系及其与LATS2 mRNA表达的关系。结果口腔鳞癌组织及癌旁正常组织LATS2 mRNA阳性表达率分别为47. 22%(51/108)、100%(45/45),LATS2 mRNA相对表达量分别为0. 32±0. 04、1. 01±0. 08,二者比较P均<0. 05。口腔鳞癌组织LATS2基因启动子完全甲基化率高于癌旁正常组织,完全非甲基化率低于癌旁正常组织,二者比较P均<0. 05。口腔鳞癌组织LATS2基因启动子甲基化与肿瘤临床分期、组织分级及淋巴结转移有关,并与LATS2 mRNA表达呈负相关关系(P均<0. 05)。结论 LATS2基因启动子甲基化可能通过导致LATS2基因表达下调,促进口腔鳞癌的发生、发展。Objective To study the promoter methylation level of large tumor suppressor homolog 2(LATS2)gene in oral squamous cell carcinoma(OSCC)tissues and its clinical significance.Methods Totally 108 cases of OSCC tissue specimens(OSCC group)and 45 cases of adjacent normal tissues(>2 cm distance from OSCC tissues,the control group)were chosen.The mRNA expression of LATS2 was analyzed by reverse transcription-polymerase chain reaction.The methy-lation specific polymerase chain reaction was used to detect the promoter methylation level of LATS2 gene.The relation be-tween promoter methylation and gene expression of LATS2 was detected by Spearman method.Results The positive ex-pression rates of LATS2 mRNA in the OSCC group and control group were 47.22%(51/108)and 100%(45/45),re-spectively;the relative expression levels of LATS2 mRNA were 0.32±0.04 and 1.01±0.08,respectively(all P<0.05).The promoter exhaustive methylation rate of LATS2 in OSCC group was significantly higher than that in the control group(P<0.05),and the promoter exhaustive non-methylation rate of LATS2 in the OSCC group was significantly lower than that in the control group(P<0.05).The promoter methylation status of LATS2 in OSCC tissues was related to the clinical staging,tumor differentiation degree and lymphatic metastasis(all P<0.05).Conclusion The promoter methyl-ation of LATS2 gene may promote the occurrence and development of OSCC by decreasing LATS2 gene expression.
