摘  要:目的探讨沉默成纤维细胞生长因子4(FGF4)表达对乳腺癌细胞增殖和凋亡的影响及其机制。方法取传2代对数生长期人乳腺癌细胞MCF-7(以下称MCF-7细胞),随机分为FGF4沉默组、阴性对照组、空白对照组,FGF4沉默组、阴性对照组按脂质体LipofectamineTM2000转染试剂说明分别转染siRNA、阴性对照序列,空白对照组不予转染。收集各组转染48 h细胞,采用MTT法检测再培养24、48、72 h细胞增殖抑制率。采用流式细胞术检测转染48 h早期凋亡率。采用Western blotting法检测转染48 h cyclin D1、caspase-3蛋白表达。结果 FGF4沉默组再培养24、48、72 h时细胞增殖抑制率均明显高于阴性对照组和空白对照组(P均<0. 05),而空白对照组与阴性对照组比较差异均无统计学意义(P均> 0. 05)。FGF4沉默组早期凋亡率明显高于阴性对照组和空白对照组(P均<0. 05),而空白对照组与阴性对照组比较差异均无统计学意义(P均> 0. 05)。FGF4沉默组cyclin D1蛋白相对表达量明显低于阴性对照组和空白对照组(P均<0. 05),caspase-3蛋白相对表达量明显高于阴性对照组和空白对照组(P均<0. 05),而空白对照组与阴性对照组比较差异均无统计学意义(P均> 0. 05)。结论沉默FGF4表达可抑制乳腺癌细胞增殖并促进其凋亡,其机制可能与抑制cyclin D1、促进caspase-3蛋白表达有关。Objective To investigate the effect of silencing fibroblast growth factor 4(FGF4)expression on prolifera-tion and apoptosis of breast cancer cells and its mechanism.Methods The human breast cancer cells MCF-7(MCF-7 cells)of the second generations were randomly divided into the siRNA interference FGF4 silencing group,negative control group,and blank control group.The siRNA and negative control sequences were transfected according to the liposome Li-pofectamineTM 2000 transfection reagent,and the blank control was not transfected.The cells of the three groups transfected for 48 h were collected,and the proliferation inhibition rate of the cells at 24,48,and 72 h after the culture was detected by MTT assay.Flow cytometry was used to discover the early apoptosis rate of cells at 48 h.Western blotting was applied to explore the expression of cyclin D1 and caspase-3 of cells at 48 h after transfection.Methods After 24,48,and 72 h of cultivation in each group,the cell proliferation inhibition rate of the FGF4 silencing group was significantly higher than that of the negative control and the blank control group(P<0.05).However,there was no statistical significance between the blank control and the negative control groups(P>0.05).The early apoptotic rate of the FGF4 silencing group was out-standingly higher than that of the other two groups(P<0.05).And no statistically significant difference was found be-tween the blank control and the negative control group(P>0.05).The relative expression of cyclin D1 protein in the FGF4 silencing group was obviously lower than that of the negative control and blank control groups(P<0.05),and the relative expression of caspase-3 protein increased remarkably as compared with that of the other groups,but no statistical difference was found between the negative control group and the blank control group(P>0.05).Conclusion Silencing FGF4 expression can inhibit breast cancer cell proliferation and promote its apoptosis,and its mechanism may be related to the inhibition of cyclin D1 and promoti
