摘 要:目的观察香芹酚(carvacrol,CAR)对糖尿病小鼠主动脉血管重构的影响。方法腹腔注射链脲佐菌素(streptozotocin,STZ)建立1型糖尿病C57BL/6小鼠模型,成模小鼠随机分为糖尿病组、CAR低、高剂量组(10、20 mg·kg^(-1)),以同周龄、同性别正常小鼠为对照组。糖尿病组和对照组每天腹腔注射生理盐水,CAR给药组每天腹腔注射CAR,每周测量体质量和随机血糖。给药6周后处死小鼠,收集主动脉,检测主动脉血管形态,细胞表型转变标志物α-actin、Ki67的表达,以及PDK1、Akt的磷酸化水平。结果 CAR可明显降低糖尿病小鼠的随机血糖,增加糖尿病小鼠主动脉Ki67的表达,以及PDK1、Akt的磷酸化水平,抑制α-actin的表达。结论CAR可降低1型糖尿病小鼠血糖,改善主动脉血管重构,对糖尿病及其并发的血管疾病有一定的治疗作用。To investigate the effect of carvacrol on aortic vascular remodeling in diabetic mice.Methods Diabetes was induced by intraperitoneal injection of streptozotocin into male mice at the dose of 45 mg·kg-1 body weight.Mice were randomly divided into three groups and age matched male C57BL/6 mice were used as normal control.The mice in diabetic group and control group were intraperitoneally injected with physiological saline once daily.The mice in carvacrol-administered group were intraperitoneally injected with carvacrol at the dose of 10 or 20 mg·kg-1 body weight respectively once daily.The body weight and random blood glucose were measured weekly.The mice were sacrificed after six weeks of administration,and the aortas were collected for detection of morphology,expression ofα-actin and Ki67,and phosphorylation levels of PDK1 and Akt.Results Carvacrol significantly reduced the random blood glucose of diabetic mice,increased the expression of Ki67 and the phosphorylation levels of PDK1 and Akt,whereas it inhibited the expression ofα-actin in aorta of diabetic mice.Conclusions Carvacrol has anti-diabetic property and can improve aortic vascular remodeling,which has the potential for development into a drug to prevent hyperglycemia and its complications of vascular diseases.
