摘 要:目的本实验研究β-淀粉样蛋白(β-Amyloid peptide,Aβ)诱导小胶质细胞产生氧化应激过程中信号转导与转录激活子3 (signal transducer and activator of transcription 3,STAT3)的作用。方法用siRNA沉默STAT3的表达后,实验分为正常对照组、Aβ处理组、Control siRNA+Aβ组以及siRNA STAT3+Aβ组。用活性氧(reactive oxygen species,ROS)检测试剂盒测定细胞内ROS水平;用生物化学方法检测一氧化氮(nitric oxide,NO)含量。结果小胶质细胞中氧化应激水平的变化:与正常对照组相比,Aβ处理组小胶质细胞中ROS水平和NO含量均显著增加;与Aβ处理组相比,siRNA STAT3+Aβ组细胞中ROS水平和NO含量均显著降低。结论 siRNA沉默BV2小胶质细胞中STAT3的表达后,可减弱Aβ寡聚体引起的小胶质细胞氧化应激的水平,STAT3可能参与调控Aβ寡聚体诱导的小胶质细胞产生氧化应激的过程。Objective In this study,we investigated the function of signal transducer and activator of transcription 3(STAT3)in the oxidative stress of microglia induced by beta amyloid peptide.Methods After the expression of STAT3 was silent with siRNA,the the experimental groups were divided into normal control,Aβgroup,control siRNA+Aβ,and siRNA STAT3+Aβ.Then the level of intracellular reactive oxygen species(ROS)was detected with ROS detection kit;Biochemical method was used for detection detecting the contents of nitric oxide(NO).Results The changes of oxidative stress in microglia:BV2 cells stimulation with Aβled to a gradual increase in the levels of ROS and NO compared with normal control group.While pretreatment with siSTAT3 significantly inhibited the Aβ-induced the production of ROS and NO compared with Aβgroup.Conclusion Supression of STAT3 using siRNA can reduce oxidative stress in microglial cells induced by Aβoligomers.STAT3 may be involved in the regulation of the process of oxidative stress in microglia induced by Aβoligomers.
