摘 要:目的观察二甲双胍对人肝癌细胞HepG_2白细胞抗原Ⅰ类分子(HLA-Ⅰ)表达的影响。方法HepG_2常规培养后给予不同浓度二甲双胍干预,分为对照组及二甲双胍处理组(0.5、1、2、5、10 mmol/L),采用实时荧光定量PCR检测HLA-Ⅰα链(HLA-E、HLA-A2402)和β链(β2-M)mRNA的表达;并采用流式细胞技术检测HepG_2细胞膜表面HLA-Ⅰ蛋白的情况。结果与对照组相比,qPCR发现5、10 mmol/L二甲双胍可显著提高HepG_2中HLA-A2402、HLA-E和β2-M mRNA表达水平(P <0.05);同时发现不同浓度二甲双胍处理并不影响HepG_2细胞膜表面HLA-Ⅰ蛋白的表达。结论二甲双胍能显著提高肝癌细胞中HLA-Ⅰ mRNA的表达水平,将为二甲双胍抗肿瘤治疗等提供依据。Objective To observe the effect of metformin on the expression of HLA-I in human hepatocel-lular carcinoma cell line HepG2.Methods HepG2 were cultured routinely and incubated then with metformin at different concentrations of metformin(0.5,1,2,5 and 10 mmol/L).HLA-Iαchain(HLA-E,HLA-A2402)andβ-chain(β2-M)mRNA were assayed with RT-PCR.The level of HLA-I protein on the surface of HepG2 was detected by flow cytometry.Results Compared with the control group,real-time quantitative PCR showed that 5 mmol/L and 10 mmol/L metformin significantly enhanced the expression of HLA-A2402,HLA-E andβ2-M mRNA in HepG2(P<0.05).The concentration of metformin did not affect the expression of HLA-I on the membrane surface of HepG2.Conclusion Metformin can significantly increase the expression of HLA-I mRNA in hepatocel-lular carcinoma cells and provide evidence for the anti-tumor therapy of metformin.
