摘 要:目的研究骨形成蛋白7 (BMP-7)对人牙髓细胞(h DPCs)增殖和分化的影响。方法原代培养h DPCs并鉴定,采用不同浓度的BMP-7处理h DPCs,检测各处理组细胞增殖活性、钙化结节量、碱性磷酸酶(ALP)活性,以及牙本质基质蛋白(DMP-1)和牙本质涎磷蛋白(DSPP)等成牙本质相关基因的表达。结果 BMP-7对h DPCs增殖无明显影响,但可剂量依赖性地促进钙化结节的形成,提高细胞中ALP活性,并上调DMP-1和DSPP的基因表达水平。结论BMP-7对h DPCs增殖无促进作用,但可明显促进h DPCs向成牙本质细胞分化。Objective To study the effects of bone morphogenetic protein-7(BMP-7)on the proliferation and odontoblast differentiation of human dental pulp cells(hDPCs).Methods Human DPCs were primary cultured and identified.Cellular proliferation,the quantity of calcified nodules,ALP activity,and the mRNA expression of dentin matrix protein 1(DMP-1)and dentin sialophosphoprotein(DSPP)in hDPCs were detected after treatment with different concentrations of BMP-7.Results BMP-7 had no significant effect on hDPCs proliferation,but could increase the mineralized nodules,ALP activity,and the expression of DMP-1 and DSPP mRNA in a dose-dependent manner.Conclusion MP-7 has no effect on the proliferation of hDPCs,but it can significantly promote the differentiation of hDPCs into odontoblasts.
