摘  要:目的探讨β-隐黄素对实验性牙周炎大鼠氧化应激的影响及可能的作用机制。方法选取30只雄性SD大鼠随机分为3组即正常对照组、牙周炎组、β-隐黄素治疗组。采用正畸结扎丝结扎双侧上颌第二磨牙颈部联合注射内毒素(LPS)的方法建立牙周炎动物模型。β-隐黄素给药7 d后,行腹主动脉采血后处死。分离双侧上颌骨,左侧用于组织学分析各组大鼠牙周组织炎症和牙槽骨丧失的情况,右侧分离牙龈组织并检测牙龈和血清中的丙二醛(MDA)的含量以及超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSHPx)的活性。结果与牙周炎组比较,β-隐黄素治疗组炎症细胞浸润数和牙槽骨丧失量减少,牙龈组织和血清中MDA含量明显降低(P<0.05),SOD和GSH-Px活性明显增高(P<0.05)。结论β-隐黄素可以减轻实验性牙周炎大鼠的氧化应激反应。Objective To investigate the effect ofβ-cryptoxanthin on oxidative stress in experimental periodontitis rats and its possible mechanism.Methods Thirty male Sprague Dawley rats were divided randomly into three groups:normal,periodontitis,periodontitis treated withβ-cryptoxanthin.Ligation with steel wire around bilateral maxillary second molar cervix and local injection with lipopolysaccharide(LPS)were used to induce experimental periodontitis model.After the rats were administrated withβ-cryptoxanthin for seven days,blood samples were collected from abdominal aorta,and then all subjects were executed.The bilateral maxilla were separated and the left side was used for histopathology analysis of inflammatory cells and alveolar bone loss(ABL).The gingival tissue of right side was separated and serum samples were collected,and then these samples were used to detect the content of malondialdehyde(MDA)and the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px).Results Compared with the periodontitis group,the number of inflammatory cells infiltration and alveolar bone loss were significantly lower in theβ-cryptoxanthin treatment group.In addition,the MDA gingival tissues and serum levels significantly decreased(P<0.05),and the activity of SOD and GSH-Px increased(P<0.05).Conclusion Treatment withβ-cryptoxanthin can relieve oxidative stress in experimental periodontitis rats.
