摘  要:研究去甲肾上腺素转运蛋白(Norepinephrine Transporter,NET)在结肠癌中的作用。利用TCGA数据库分析NET与临床结肠癌患者的TNM及stage分期的关系;收集临床标本行NET免疫组化实验;在结肠癌细胞中下调NET的表达行Transwell实验和Western Blot观察其变化。临床生物大数据及临床标本免疫组化结果显示:NET与结肠癌患者的转移及分期呈正相关,下调NET的表达显著抑制人结肠癌细胞的迁移能力以及迁移相关分子MMP9的表达,去甲肾上腺素转运蛋白参与对人结肠癌细胞迁移能力的调控。To explore the role of norepinephrine transporter(NET)in colon cancer.The relationship between NET and TNM and stage of patients with colon cancer was analyzed using TCGA database.The clinical samples of colon cancer were collected and prepared for immunohistochemistry assay of NET.Changes of migration and proteins were detected by Transwell assay and Western Blot in NET-downregulated human colon cancer cells.Clinical biologic data and immunohistochemical results of clinical specimens showed a positive correlation between NET and metastasis and staging of colon cancer patients.Down-regulation of NET expression significantly inhibited the migration of human colon cancer cells and the expression of the migration-associated molecule MMP9.NET might play a critical role in the regulation of migration ability of human colon cancer cells.
