摘 要:目的观察淫羊藿苷(ICA)预处理对急性肝损伤大鼠肝功能的保护作用,并探讨其作用机制。方法将40只大鼠随机分为对照组、模型组、ICA低剂量组、ICA高剂量组各10只,ICA低、高剂量组分别给予腹腔注射ICA 30、80 mg/kg,对照组和模型组给予等量生理盐水,1次/d,连续7 d。末次给药后2 h,模型组和ICA低、高剂量组给予0. 35%CCl4灌胃制作肝损伤模型,对照组给予等量生理盐水灌胃,12 h后处死。取大鼠股动脉血,采用生化分析仪检测丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST)活性;取肝组织,采用考马斯亮蓝法检测超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)水平,ELISA法检测肿瘤坏死因子α(TNF-α)水平。结果与对照组比较,模型组血清ALT、AST活性升高,肝组织SOD、GSH-Px活性降低,MDA、TNF-α水平升高(P均<0. 01);与模型组比较,ICA低剂量组、ICA高剂量组血清ALT、AST活性降低,肝组织SOD、GSH-Px活性增高,MDA、TNF-α水平降低(P均<0. 01)。结论 ICA对急性肝损伤大鼠的肝功能具有保护作用,其机制可能与增加SOD、GSH-Px活性,降低MDA、TNF-α水平,从而减轻自由基损害和炎症反应有关。Objective To explore the protective effect and mechanism of icariin(ICA)on the liver function of rats with acute liver injury.Methods Forty SPF rats were randomly divided into four groups:the control group,model group,low-dose ICA group,and high-dose ICA group.The rats in the low-dose ICA and high-dose ICA group were injected intraperitoneally with ICA 30 and 80 mg/kg.The rats in the control group and model group were given the same amount of normal saline once daily for 7 days.Two hours after the last administration,the model group and the low-dose ICA group and high-dose ICA group were given 0.35%CCl 4 to prepare a liver injury model.The control group was given the same amount of normal saline and was sacrificed 12 hours later.Rat femoral artery blood was obtained.The alanine aminotransferase(ALT)activity,aspartate transaminase(AST)activity,superoxide dismutase(SOD)activity,glutathione peroxidase(GSH-Px)activity and malondialdehyde(MDA)content in rats were measured;tumor necrosis factor-α(TNF-α)content in rats were detected by ELISA.Results Compared with the control group,the ALT activity and AST activity significantly increased,while SOD activity,GSH-Px activity,MDA content,and TNF-αcontent significantly decreased in the model group(all P<0.01).Compared with the model group,the ALT activity and AST activity significantly decreased,while SOD activity,GSH-Px activity,MDA content,and TNF-αcontent significantly increased in the low-dose ICA group and high-dose ICA group(all P<0.01).Conclusion ICA exerts the protective effect on the liver function of rats with acute liver injury induced by carbon tetrachloride through increasing the SOD activity and GSH-Px activity,decreasing MDA and TNF-αlevels,thereby reducing free radical damage and inflammatory response.
