摘  要:目的建立一种基于导电聚吡咯和纳米金复合电极的牙周细菌检测电化学免疫传感器。方法通过电化学聚合方法,在金叉指微电极表面修饰聚吡咯阵列/纳米金颗粒;在修饰微电极表面固定牙龈卟啉单胞菌多抗;利用扫描电镜和拉曼光谱对电极表面修饰过程进行表征;通过阻抗-细菌浓度函数关系分析传感器的线性检测范围和重现性。结果恒电流法在金叉指电极表面成功获得聚吡咯纳米线,平均直径约80 nm;聚合电流密度10μA,聚合时间10 min;循环伏安法在聚吡咯表面成功沉积纳米金颗粒,循环电压从-0. 1到1. 5 V,循环速度50 m V/s,循环10次;传感器线性检测范围为103-109cells/ml,重现性好,检测时间在1 h内。结论基于导电聚吡咯和纳米金复合电极的牙周细菌检测微传感器在一定范围内能够用于牙龈卟啉单胞菌的快速量化。Objective To develop an electrochemical immunosensor based on polypyrrole nanowires array/gold nanoparticles modified microelectrodes for quantifying periodontal bacteria.Methods By electrochemical polymerized method,polypyrrole/Au nanoparticles modified microelectrodes were obtained.Anti-Porphyromonas gingivalis antibodies were immobilized on composed modified microelectrodes.Scanning electron microscope and Raman spectroscopy were used to characterize the modification of microelectrode.The function relationship of impedance and bacteria concentration was used to analyze the linear detection range and reproducibility of the developed biosensor.Results The polypyrrole nanowires array was polymerized by chronopotentiometry method with the average diameter of about 80 nm,the polymerization current density was 10μA,and the polymerization time was 10 min.And then gold nanoparticles were deposited on the surface of polypyrrole array using cyclic voltammetry(CV)with potential scanning from-0.1 to 1.5 V at a scan rate of 50 mV/s for 10 cycles.The linear detection range was 10 3-10 9 cells/ml.It also had a good reproducibility and the detection time was 1 h.Conclusion The results clearly suggest that the developed immunsensor may have the potential application value for periodontal bacteria detection in clinical samples.
