摘 要:目的探讨转化生长因子β受体Ⅱ(TGFβRⅡ)在棘皮动物微管相关蛋白4-间变性淋巴瘤激酶(EML4-ALK)融合基因阳性肺腺癌患者肿瘤组织中的表达及对人EML4-ALK阳性肺腺癌细胞H3122侵袭力的影响。方法纳入EML4-ALK阳性肺腺癌患者18例,其中ⅠA期11例,ⅢA期7例,采用实时荧光定量聚合酶链反应(RT-qPCR)检测患者肺癌组织中TGFβRⅡmRNA的表达量。构建TGFβRⅡ沉默质粒(TGFβRⅡ-shRNA)及阴性对照(NC-shRNA)质粒,进行慢病毒包装后转染H3122细胞,并将转染TGFβRⅡ-shRNA、NC-shRNA质粒的细胞设为LV-shRNA-TGFβRⅡ-H3122组、LV-shRNA-GP-H3122组,测定两组细胞的侵袭能力。结果ⅠA期与ⅢA期肺腺癌组织的TGFβRⅡmRNA相对表达量差异无统计学意义(P>0. 05)。LV-shRNA-TGFβRⅡ-H3122组通过Transwell的细胞数多于LV-shRNA-GP-H3122组(P <0. 05)。结论 TGFβRⅡmRNA在早期与中晚期EML4-ALK阳性肺腺癌组织中的表达无明显差异,但沉默TGFβRⅡ基因后EML4-ALK阳性肺腺癌细胞侵袭性明显增强。Objective To explore the expression of transforming growth factorβreceptorⅡ(TGFβRⅡ)in cancer tissues from lung adenocarcinoma patients with echinoderm microtubule-associated protein-like 4-anaplastic lymphoma kinase(EML4-ALK)fusion gene positive and its effect on the invasion of lung adenocarcinoma H3122 cells with human EML4-ALK positive.Methods Real-time quantitative PCR was used to detect the expression level of TGFβRⅡmRNA in cancer tissues from 18 lung adenocarcinoma patients with EML4-ALK positive,including 11 of stageⅠA and 7 of stageⅢA.TGFβRⅡsilencing plasmids(TGFβRⅡ-shRNA)and negative control plasmids(NC-shRNA)were constructed to receive lentivirus packaging,then were transfected into H3122 cells separately,and the H3122 cells transfected by TGFβRⅡ-shRNA and NC-shRNA plasmids were enrolled as LV-shRNA-TGFβRⅡ-H3122 group and LV-shRNA-GP-H3122 group respectively.The cell invasion of the two groups was determined.Results There was no statistically significant difference in the relative expression level of TGFβRⅡmRNA between lung adenocarcinoma of stageⅠA and stageⅢA(P>0.05).The cells passing through Transwell in the LV-shRNA-TGFβRⅡ-H3122 group were more than those in the LV-shRNA-GP-H3122 group(P<0.05).Conclusion No significant difference is observed in the expression of TGFβRⅡmRNA in cancer tissues between early and advanced lung adenocarcinoma with EML4-ALK positive.However,fairly stronger invasion of lung adenocarcinoma cells with EML4-ALK positive is seen after silencing TGFβRⅡgene.
