中国福建东南部地区阿尔茨海默病患者载脂蛋白E基因型与血清生化指标相关性分析

摘　　要：目的:研究中国福建东南部地区阿尔茨海默病(Alzheimer’s disease,AD)患者载脂蛋白E(apolipoprotein E,ApoE)基因型分布,及其与血清总胆固醇、总甘油三酯和血糖关联性。方法:选取2016年3月-2017年9月期间在厦门大学附属第一医院神经内科门诊就诊的AD患者106例作为研究对象,通过临床资料分析和评估,提取其外周血DNA运用多重突变扩增系统检测ApoE基因型,同时对ApoE基因型与血清生化指标进行相关性分析。结果:中国福建东南部地区AD患者ApoE基因型中,无ApoEε2ε2,ApoEε2ε3有6例(5.66%),ApoEε2ε4有4例(3.77%),ApoEε3ε3有34例(32.08%),ApoEε3ε4有42例(39.62%),ApoEε4ε4有20例(18.87%)。在血清生化指标相关分析中,携带ApoEε4组中的血清总胆固醇水平明显高于不携带ApoEε4组,差异有统计学意义(t=2.113,P=0.038)。而两组的血清总甘油三酯和空腹血糖水平比较,差异均无统计学意义(P>0.05)。结论:中国福建东南部地区AD患者携带ApoEε4血清总胆固醇水平高于不携带ApoEε4,而总甘油三酯水平和空腹血糖水平无明显差异。Objective:To analyze the apolipoprotein E(ApoE)genotypes distribution of patients with Alzheimer’s disease(AD)in southeastern Fujian of China,and clarify the correlation between ApoE genotypes and serum total cholesterol,total triglycerides and blood glucose.Method:A total of 106 AD patients from March 2016 to September 2017 in the Neurology Clinic of the First Affiliated Hospital of Xiamen University were selected as the study objects.The clinical data of patients were analyzed and evaluated,DNA were extracted from peripheral blood and ApoE genotypes were detected by multiplex amplification refractory mutation system polymerase chain reaction.The correlation between ApoE genotypes and serum biochemical parameters were also studied.Result:Among the ApoE genotypes of AD patients in southeastern Fujian of China,there were no ApoEε2ε2,6 cases(5.66%)ApoEε2ε3,4 cases(3.77%)ApoEε2ε4,34 cases(32.08%)ApoEε3ε3,42 cases(39.62%)ApoEε3ε4 and 20 cases(18.87%)ApoEε4ε4.In the correlation analysis of serum biochemical parameters,the serum total cholesterol level of the ApoE4ε4 group was significantly higher than that of the without ApoEε4 group(t=2.113,P=0.038).There were no significant differences in the levels of serum total triglycerides and fasting blood glucose between the two groups(P>0.05).Conclusion:The serum total cholesterol levels of AD patients with ApoEε4 were higher than those without ApoEε4 in southeastern Fujian of China,but there were no significant differences between the levels of total triglycerides and fasting blood glucose.
