摘  要:目的探讨丰富环境(EE)干预对帕金森病(PD)大鼠中脑miR-133b表达的影响。方法将造模成功的6-羟基多巴胺(6-OHDA)PD大鼠随机分成2组:PD+EE组、PD+标准环境(SE)组。设假手术(sham)+EE组、sham+SE组为对照组。予对应环境干预6周后,用酪氨酸羟化酶(TH)免疫组化法检测大鼠中脑TH阳性纤维及TH阳性细胞表达;采用实时荧光定量PCR检测大鼠中脑miR-133b表达水平。结果与sham+SE组相比,PD+SE组损毁侧中脑TH阳性纤维及细胞数目较少;与PD+SE组相比,PD+EE组大鼠损毁侧中脑TH阳性纤维及细胞数目较多,差异均有统计学意义(P <0.05)。PD+SE组大鼠中脑miR-133b相对表达量高于sham+SE组;PD+EE组大鼠中脑miR-133b的相对表达量低于PD+SE组,差异均有统计学意义(P <0.05)。结论 6-OHDA可能通过上调miR-133b表达导致多巴胺能神经元退行性变。EE可能通过抑制miR-133b的表达,促进黑质-纹状体系统多巴胺能神经元的恢复。Objective To investigate the effect of the enriched environment(EE)on miR-133b expres-sion in the midbrain of a rat model of Parkinson′s disease(PD).Methods PD rats induced by 6-hydroxydopa-mine(6-OHDA)were divided into 2 groups:PD+EE group and PD+standard environment(SE)group.Rats received sham-operation were divided into 2 groups:sham+EE group and sham+SE group.Rats were exposed to different environments for 6 weeks.Tyrosine hydroxylase(TH)-positive fibers or cells in lesioned midbrain were examined by immunohistochemistry,and the relative expression of miR-133b was detected by fluorescence real-time quantitative PCR.Results Compared with PD+SE group,TH-positive fibers and cells were increased in PD+EE group,but miR-133b expression was decreased in the lesioned midbrain.Conclusion 6-OHDA may lead to neuronal degeneration by up-regulating miR-133b.EE may promote the recovery of nigrostriatal dopaminergic neu-rons through inhibiting miR-133b expression in midbrain of PD rats.
