摘 要:使用预实验筛选出的X-5型大孔树脂,对粘质沙雷氏菌产灵菌红素的发酵与分离工艺条件进行优选,结果发现,X-5型大孔吸附树脂在粘质沙雷氏菌接种发酵培养基5 h后添加60 g L^(-1)树脂继续发酵36 h,灵菌红素产量可提高23. 12%.其分离优化工艺条件是径高比为1∶4,30%乙醇洗脱体积流量为1 BV·h^(-1),洗脱体积为3 BV; 95%乙醇洗脱体积流量为1. 5 BV·h^(-1),洗脱体积为3 BV.制成的灵菌红素质量分数为(40. 37±0. 64)%,回收率为(79. 63±0. 94)%.该方法为进一步探索灵菌红素的发酵与分离耦合、提高灵菌红素产量与回收率提供了基础数据,对推动灵菌红素工业化进程具有重要意义.By using prescreened X-5 macroporous resin,the fermentation and separation conditions for Serratia marcescens to produce prodigiosin are optimized.The results show that if 60 gL^-1 resin is added five hours after the fermentation medium is inoculated with S.Serratia and then the fermentation process continues for 36 hours,the yield of prodigiosin by X-5 macroporous resin will increase by 23.12%.The optimal separation conditions are as follows:the ratio of diameter to height is 1∶4;30%ethanol elution volume flow rate is 1 BVh^-1 and elution volume is 3 BV;95%ethanol elution volume flow rate is 1.5 BVh^-1 and elution volume is 3 BV.The content of prodigiosin is(40.37±0.64)%,and the recovery rate is(79.63±0.94)%.This method provides basic data for the further exploration of the coupling of the fermentation and separation of prodigiosin and the improvement of its yield and recovery rate,and is of great importance for promoting the industrialization process of producing prodigiosin.
