摘 要:目的观察姜黄素灌胃对动脉粥样硬化(AS)大鼠血管内皮损伤的干预效果,并探讨其机制。方法 32只雄性SD大鼠适应性饲养1周后,随机均分为对照组、模型组、姜黄素组、姜黄素+Zn PPⅨ组,后三组予以高脂饲料喂养+液氮冻伤右侧颈总动脉+腹腔注射维生素D3+免疫损伤的方法制备AS模型。第14周开始药物干预,姜黄素组给予姜黄素100 mg/(kg·d)灌胃;姜黄素+Zn PPⅨ组给予姜黄素100 mg/(kg·d)灌胃+Zn PPⅨ7. 5 mg/kg隔日1次腹腔注射;模型组及对照组给予2 m L生理盐水灌胃和1 m L生理盐水腹腔注射。实验期间模型组、姜黄素组、姜黄素+Zn PPⅨ组高脂饲料喂养,对照组普通饲料喂养。干预2周,取血检测炎症指标[肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)]、氧化应激指标[丙二醛(MDA)及总抗氧化能力(T-AOC)]及血管内皮功能损伤指标[NO水平和循环内皮细胞数量];处死大鼠,取右侧颈总动脉行组织病理学观察,采用免疫组化法检测HO-1蛋白表达。结果与对照组相比,模型组颈动脉有明显AS斑块形成,血清TNF-α、IL-6及MDA水平和循环内皮细胞数量均升高,T-AOC及NO水平均降低(P均<0. 05)。与模型组相比,姜黄素组血清TNF-α、IL-6及MDA水平和循环内皮细胞数量均降低,T-AOC、NO水平及血管组织内HO-1表达均升高(P均<0. 05);姜黄素+Zn PPⅨ组血管组织内HO-1表达亦升高(P <0. 05),但血清TNF-α、IL-6、T-AOC、MDA、NO水平和循环内皮细胞数量均无明显改变(P均> 0. 05)。结论姜黄素可通过诱导HO-1蛋白表达有效抑制AS大鼠的炎症与氧化应激水平,减轻血管内皮损伤,保护内皮功能。Objective To investigate the intervention effect and mechanism of curcumin on vascular endothelial injury in rats with atherosclerosis(AS).Methods Thirty-two male SD rats were adaptively raised for 1 week,after that,they were randomly divided into 4 groups:the control group,the model group,the curcumin group,and the curcumin+ZnPPⅨgroup.The latter three groups were fed with high-fat diet along with liquid nitrogen frostbite of the right common carotid artery,intraperitoneal injection of vitamin D 3,and the immune damage in order to make the atherosclerosis models.From the fourteenth week on,drug intervention was conducted for two weeks in each group,among which,the curcumin group was given curcumin 100 mg/kg·d by gavage,the curcumin+ZnPPⅨgroup was administered curcumin 100 mg/kg·d by gavage,followed by the intraperitoneal injection of ZnPPⅨ7.5mg/kg for once every other day,while the model group was administered equal amount of normal saline.The control group was fed with normal diet and treated with the same method as that of the other three groups with normal saline instead of various drugs.The blood samples were taken to detect the inflammation indexes[tumor necrosis factor-α(TNF-α)and IL-6],oxidative stress indexes[malondialdehyde(MDA)and total antioxidant capacity(T-AOC)]and indexes for injuried function of endothelia(NO content and the amount of circulating endothelial cells in peripheral blood).The right common carotid artery was used for histopathologic detection,and the immunohistochemical analysis was used to detect the HO-1 protein expression.Results Compared with the control group,the serum levels of TNFα,IL-6 and MDA and the number of circulating endothelial cells in the model group increased significantly,while the contents of T-AOC and NO significantly decreased(all P<0.05),accompanied by significant AS plaques formation in the carotid artery.Compared with the model group,the expression of HO-1 protein in the vessel of the curcumin group increased significantly,the levels of serum TNFα,IL
