胃腺癌组织中TM4SF5 mRNA、蛋白的表达变化及其意义

摘　　要：目的观察胃腺癌组织中四次跨膜蛋白超家族5(TM4SF5) mRNA、蛋白的表达变化,并探讨其意义。方法选取胃腺癌组织及癌旁正常组织各76例份,采用qRT-PCR法和免疫组化法分别检测其TM4SF5 mRNA、蛋白,并分析TM4SF5蛋白表达与胃腺癌临床病理参数的关系。结果胃腺癌组织、癌旁组织中TM4SF5 mRNA相对表达量分别为1. 236±0. 731、0. 402±0. 210,两者比较,P <0. 05。胃腺癌、正常组织中TM4SF5蛋白阳性率分别为59. 2%(45/76)、13. 2%(10/76),两者比较,P <0. 05。TM4SF5蛋白表达与胃腺癌淋巴结转移、浸润深度相关(P均<0. 05)。结论胃腺癌组织中TM4SF5 mRNA、蛋白表达升高,TM4SF5蛋白表达与胃腺癌淋巴结转移、浸润深度相关,其可能与胃腺癌的发生发展有关。To observe the expression changes of TM4SF5 mRNA and protein in the gastric adenocarcinoma tissues,and to investigate their significance.Methods Seventy-six cases of gastric adenocarcinoma tissues and normal adjacent tissues were selected.TM4SF5 mRNA was detected by qRT-PCR,and TM4SF5 protein was detected by immunohistochemistry.Meanwhile,the relationship between TM4SF5 protein expression and clinical pathological parameters of gastric adenocarcinoma was analyzed.Results The relative expression of TM4SF5 mRNA in the gastric adenocarcinoma tissues and adjacent tissues was 1.236±0.731 and 0.402±0.210,respectively(P<0.05).The positive rate of TM4SF5 protein in the gastric adenocarcinoma and normal tissues was 59.2%(45/76)and 13.2%(10/76),respectively(P<0.05).TM4SF5 protein expression was correlated with lymph node metastasis and depth of invasion(both P<0.05).Conclusion TM4SF5 mRNA and protein expression increases in the gastric adenocarcinoma tissues,and TM4SF5 protein expression is related to lymph node metastasis and depth of invasion of gastric adenocarcinoma,which may be related to the occurrence and development of gastric adenocarcinoma.
