摘 要:目的观察GLI1表达与结肠癌上皮-间质转换的关系。方法收集江西省中西医结合医院2010年1月~2015年12月诊断明确、资料完整的50例手术切除结肠癌标本及其癌旁组织,做成组织芯片,采用免疫组化SP的方法检测组织芯片中结肠癌及癌旁组织中GLI1及Vimentin、N-cadherin、E-cadherin表达。结果 GLI1在结肠癌中的表达率高于癌旁组织,差异有统计学意义(P<0.05),与结肠癌的淋巴转移、临床分期、肿瘤分化明显有关(P<0.05);E-cadherin在癌旁组织中的表达率高于结肠癌,差异有统计学意义(P<0.05),与结肠癌的淋巴转移、临床分期、肿瘤分化明显有关(P<0.05);Vimentin、N-cadherin在结肠癌中的表达率高于癌旁组织,差异有统计学意义(P<0.05),与结肠癌的淋巴转移、临床分期、肿瘤分化明显有关(P<0.05)。GLI1表达与上皮-间质转换的状态有关(P<0.05)。结论结肠癌中GLI1的表达与Vimentin、N-cadherin、E-cadherin有关,提示GLI1有可能参与了结肠的上皮-间质转化过程。Objective To observe the relationship between the expression of GLI1 and epithelial-mesenchymal transformation in colon cancer.Methods From January 2010 to December 2015,altogether 50 specimens of surgically resected colon cancer and adjacent tissues from Jiangxi Integrated Hospital of Traditional Chinese and Western Medicine were collected and made into tissue microarrays.Immunohistochemical SP method was used to detect the expression of GLI1,Vimentin,N-cadherin and E-cadherin in colon cancer tissues and adjacent tissues.Results The expression of GLI1 in the colon cancer was higher than that in the adjacent tissues,with statistically significant difference(P<0.05),which was significantly correlated with lymphatic metastasis,clinical stage and tumor differentiation of colon cancer(P<0.05).The expression of E-cadherin in the adjacent tissues was higher than that in the colon cancer,with statistically significant difference(P<0.05),which was significantly correlated with lymphatic metastasis,clinical stage and tumor differentiation of colon cancer(P<0.05).The expression of vimentin and N-cadherin were higher in the colon cancer than that in the adjacent tissues,with statistically significant difference(P<0.05),which was significantly associated with lymphatic metastasis,clinical stage,and tumor differentiation of colon cancer(P<0.05).The expression of GLI1 was associated with the state of epithelial-mesenchymal transformation(P<0.05).Conclusion The expression of GLI1 in the colon cancer is related to Vimentin,N-cadherin and E-cadherin.It is indicated that GLI1 may participate in the epithelial-mesenchymal-transformation process.
