摘 要:目的检测miR-489对黑色素瘤细胞增殖的影响。方法在黑色素瘤细胞A375中分别过表达或沉默miR-489后,通过MTT实验观察miR-489对细胞增殖的影响。进一步通过Western blotting和实时PCR检测miR-489对A375细胞中增殖相关蛋白CDK2和cyclin E的影响。结果 MTT检测发现,miR-489过表达后可以显著抑制A375细胞增殖,miR-489受到抑制后A375细胞的增殖受到促进。Western blotting和实时PCR检测发现,过表达miR-489可以抑制A375细胞中增殖相关蛋白CDK2和cyclin E蛋白的表达;miR-489受到抑制后蛋白表达增加。结论 miR-489对黑色素瘤细胞的增殖有一定的抑制作用,可能作为黑色素瘤诊疗的新靶点。Objective To determine the effect of miR-489 on the proliferation of melanoma cells.Methods MTT assay was used to observe the effect of miR-489 on the proliferation of A375 cells after miR-489 was overexpressed or silenced.Western blotting and real-time PCR were used to determine the effect of miR-489 on the expression of CDK2 and cyclin E.Results MTT assays showed that overexpression of miR-489 significantly inhibited the proliferation of A375 cells and that silencing miR-489 promoted the proliferation.Western blotting and real-time PCR showed that overexpression of miR-489 inhibited the expression of CDK2 and cyclin E in A375 cells and that silencing miR-489 increased their expression.Conclusion miR-489 can inhibit the proliferation of melanoma cells and could be used as a new target for the diagnosis and treatment of melanoma.
