摘 要:目的研究熊果酸对大鼠急性酒精性肝损伤的保护作用及机制。方法将SD大鼠随机分为对照组、模型组、熊果酸组、联苯双酯组,模型组、熊果酸组、联苯双酯组予以50%乙醇灌胃处理,建立急性酒精性肝损伤模型,熊果酸组、联苯双酯组分别灌胃给予熊果酸60 mg/kg、联苯双酯5. 625 mg/kg,对照组和模型组给予等体积生理盐水灌胃处理。每隔3 d尾部采血检测大鼠血糖。给药14 d后,检测各组大鼠空腹血糖(FPG)、血清胰岛素(FINS)、ALT、AST、GGT、TG及肝组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量。结果与对照组比较,模型组FPG升高(P <0. 001),血清FINS降低(P <0. 001),胰岛素敏感指数(ISI)降低(P <0. 001),ALT、AST、GGT、TG均升高(P <0. 005),SOD活性降低(P <0. 05),MDA升高(P <0. 05),建模成功。与模型组比较,熊果酸组血清ALT、AST、GGT、TG及肝组织MDA水平升高(P <0. 001),SOD活性增加(P <0. 001),FPG水平下降(P <0. 001),FINS含量升高(P <0. 001)。熊果酸组各项指标与联苯二酯组比较差异无统计学意义(P均> 0. 05)。结论熊果酸对乙醇诱导的肝损伤大鼠肝功能有保护作用,其机制可能与减轻脂质过氧化、抑制氧化应激有关。Objective To study the protective effect and mechanism of ursolic acid on rats with acute alcoholic liver injury.Methods SD rats were randomly divided into the control group,model group,ursolic acid group(60 mg/kg),and bifendate group(5.625 mg/kg).Except the control group,the acute alcoholic liver injury models were established by intragastric administration of 50%alcohol.The rats in the ursolic acid group and bifendate group were treated with the corresponding drugs,while the rats in the control group and model group were administered equal volume of normal saline.We detected the blood glucose per 3 days from the tail blood.After 14 days,we detected the blood glucose,insulin,serum ALT,AST,GGT,TG,and the superoxide dismutase(SOD)activity and malondialdehyde(MDA)levels in the liver tissues.Results Compared with the control group,FPG increased(P<0.001),serum FINS decreased(P<0.001),insulin sensitive index(ISI)reduced(P<0.001),ALT,AST,GGT,TG significantly increased(P<0.005),SOD activity reduced(P<0.05),and MDA increased(P<0.05)in the model group,which indicated the successful modeling.Compared with the model group,serum ALT,AST,GGT,TG,and liver tissue MDA level,SOD activity increased(P<0.001),FPG decreased(P<0.001),and FINS content increased(P<0.001)in the ursolic acid group;no statistically significant difference was found in each index between the ursolic acid group and biphenyl ester group(all P>0.05).Conclusion Ursolic acid can protect the rats with ethanol-induced liver injury by relieving lipid peroxidation and inhibiting oxidative stress.
