胃癌组织中LncRNA NNT-AS1的表达及意义

摘　　要：目的观察胃癌组织中长链非编码RNA(LncRNA)烟酰胺核苷酸反义转氢酶RNA1(NNT-AS1)的表达并探讨其意义。方法采用实时荧光定量PCR(RT-PCR)法检测77例份胃癌组织及相应癌旁正常胃组织中LncRNA NNT-AS1的表达,并分析LncRNA NNT-AS1表达与胃癌患者临床病理特征的关系。结果胃癌及癌旁正常组织中LncRNA NNT-AS1相对表达量分别为0. 509±0. 110、0. 103±0. 004,二者比较差异有统计学意义(t=32. 366,P=0. 000)。LncRNA NNT-AS1在胃癌组织中的表达与肿瘤分化程度、TNM分期、脉管侵袭、淋巴结转移相关(P均<0. 05),而与患者年龄、性别、神经侵袭无关(P均> 0. 05)。胃癌组织中LncRNA NNT-AS1表达较低者生存率与生存时间均高于LncRNA NNT-AS1表达较高者(P均<0. 05)。结论胃癌组织中LncRNA NNT-AS1呈异常高表达,LncRNA NNT-AS1可能是胃癌发生及恶性进展的重要因素,检测LncRNA NNT-AS1表达有助于患者预后判断。Objective To observe the expression of long non-coding RNA(lncRNA)nicotinamide nucleotide transhydrogenase-antisense RNA1(NNT-AS1)in the gastric carcinoma tissues and to explore its significance.Methods We applied the real-time polymerase chain reaction(RT-PCR)to determine the expression of LncRNANNT-AS1 in the carcinoma tissues and paired adjacent normal gastric tissues from 77 cases of patients with gastric carcinoma,then we analyzed the correlation between the expression of LncRNANNT-AS1 and clinicopathological features.Results The relative expression of LncRNANNT-AS1 in the gastric carcinoma tissues and the adjacent normal gastric tissues was 0.509±0.110 and 0.103±0.004,respectively;the difference was statically significant(P<0.05).The expression of LncRNANNT-AS1 in the carcinoma tissues was related to differentiation,TNM stage,vessel invasion and lymph node metastasis(all P<0.05),but not related to age,gender or neural invasion(all P>0.05).The total survival rate and average survival time of patients with low expression of LncRNANNT-AS1 were significantly higher than those of patients with high expression(all P<0.05).Conclusions The LncRNANNT-AS1 is abnormally highly expressed in the gastric carcinoma tissues.LncRNANNT-AS1 may be an important regulatory factor in the gastric carcinogenesis and malignant development,and detection of LncRNANNT-AS1 helps to determine the prognosis of patients.
