摘 要:目的观察乳腺癌组织中Bmi1、miR-221-3p的表达变化,并探讨其临床意义。方法采用实时荧光定量PCR法检测81例份乳腺癌患者癌组织及癌旁组织中的Bmi1、miR-221-3p相对表达。分析癌组织中Bmi1、miR-221-3p表达与患者临床病理特征及预后的关系。结果乳腺癌组织、癌旁组织中Bmi1的相对表达量分别为24. 23±1. 23、1. 04±0. 12,二者相比,P <0. 05;乳腺癌组织、癌旁组织中miR-221-3p的相对表达量分别为0. 43±0. 02、1. 06±0. 23,二者相比,P <0. 05。两种组织中Bmi1与miR-221-3p表达呈负相关(r=-0. 597,P <0. 01)。乳腺癌组织中Bmi1和miR-221-3p表达与肿瘤结节数、肿瘤局部浸润、淋巴结转移及临床分期有关(P均<0. 05)。乳腺癌组织中Bmi1高表达者5年总生存率为61. 54%(24/39)、低表达者为90. 48%(38/42),二者相比,P <0. 01。miR-221-3p高表达者5年总生存率为86. 75%(37/43)、低表达者为57. 89%(22/38),二者相比,P <0. 01。结论乳腺癌组织中miRNA-221-3p表达下调,Bmi1表达上调;乳腺癌组织中miRNA-221-3p低表达和Bmi1高表达预示患者肿瘤结节较多且处于较高临床分期、淋巴结转移可能性较大,并伴有局部浸润,预后较差。Objective To observe the expression changes of Bmi1 and miR-221-3p in the breast cancer tissues and to explore its clinical significance.Methods The relative expression levels of Bmi1 and miR-221-3p in the cancer tissues and adjacent tissues form 81 patients with breast cancer were detected by real-time fluorescence quantitative PCR.The relationship between the expression of Bmi1 and miR-221-3p in cancer tissues and clinicopathological parameters and prognosis was analyzed.Results The relative expression level of Bmi1 in the breast cancer tissues and the adjacent tissues was 24.23±1.23 and1.04±0.12(P<0.05);the expression of miR-221-3p in the breast cancer tissues and adjacent tissues was 0.43±0.02 and 1.06±0.23,respectively(P<0.05).There was a negative correlation between the expression of Bmi1 and miR-221-3p(r=-0.597,P<0.01).The expression of Bmi1 and miR-221-3p in the breast cancer tissues was related to the number of tumor nodules,tumor local infiltration,lymph node metastasis,and clinical stage(all P<0.05).In breast cancer tissues,the 5-year overall survival rate in patients with high Bmi1 expression was 61.54%(24/39),and was 90.48%(38/42)in patients with low expression(P<0.01);the 5-year overall survival rates in patients with high miR-221-3p expression and low miR-221-3p expression were 86.75%(37/43)and 57.89%(22/38)with significant difference(P<0.01).Conclusion Bmi1 is highly expressed in the breast cancer tissues,and miR-221-3p is lowly expressed.The high expression of Bmi1 and the low expression of miR-221-3p in the breast cancer tissues indicate that there are more tumor nodules which are at a higher clinical stage,and with local infiltration,meanwhile,the possibility of lymph node metastasis is greater,and the prognosis is poor.
