摘 要:马疱疹病毒1型(EHV-1)可引起马属动物多系统疾病,呈世界性分布并造成严重经济损失。为进一步认识EHV-1的分子特征,采用PCR分段扩增测序,利用ORF finder和Smart BLAST分析基因组结构及基因功能,用DAN Man和MEGA7.0进行基因组序列特征及核苷酸同源性分析。结果表明,XJ2015株全基因组序列长度为150 267bp。G+C含量为56.7%。结构简式为TRL-UL-IRL-IRS-US-TRS,包含80个开放阅读框,编码78个蛋白。同源性分析表明,32株病毒全基因组核苷酸同源性为86.74%,TRL/IRL高达99.49%,TRS/IRS仅为65.99%。XJ2015株与英国株Ab4同源性最高(93.58%),与美国株T-616同源性最低(76.72%)。首次获得并分析我国流行毒株EHV-1XJ2015全基因序列,为进一步研究EHV-1的遗传背景和进化关系及基因功能的挖掘提供理论依据。Equine herpesvirus 1(EHV-1)is a pathogen of major importance in horses that can induce a wide spectrum of diseases.It distributes worldwide and causes severe economic losses.This research aimed to comprehend molecular characteristics of EHV-1.Genomic DNA were sequenced with the application of PCR.ORF finder and Smart BLAST were used for analysis of genome structure and gene function.The software of DANMAN and MEGA7.0 were used for genome characteristics and nucleotide homology analysis.The results showed that the genome of EHV-1-XJ2015 is 150 267 bp in size,has a base composition of 56.7%G+C,and contains 80 open reading frames to encode 78 proteins.Genomic structure is TR L-UL-IR L-IR S-US-TR S.Homologous analysis showed that the DNA homology of the 32 EHV-1 genomes reaches 86.74%,in which TR L/IR L reaches 99.49%and TR S/IR S only 65.99%.EHV-1-XJ2015 has highest homology with Ab4 from UK(93.58%)and the lowest homology with T-616 from USA(76.72%).In the study,it is the first report that the genome sequence of EHV-1 is obtained in China.It provides a theoretical basis for studying the genetic background,evolutionary relationship and gene function of EHV-1 in the further.
