摘  要:目的探讨尿路上皮癌相关1(UCA1)和miR-141在膀胱癌细胞和膀胱癌组织中的表达及与临床预后相关性。方法测定人膀胱上皮永生化细胞株SV-HUC-1及人膀胱癌细胞株BIU-87、EJ、T24、T24-MMC、T24-ADM UCA1和miR-141 mRNA表达情况;同时测定78例患者膀胱癌组织及癌旁正常组织UCA1、miR-141 mRNA表达水平,分析其表达情况与患者预后的相关性。结果膀胱癌细胞株BIU-87、EJ、T24、T24-MMC、T24-ADM组UCA1、miR-141mRNA水平明显高于膀胱上皮永生化细胞株SV-HUC-1组,差异有统计学意义(P <0. 05);耐药膀胱癌细胞株T24-MMC、T24-ADM组UCA1、miR-141 mRNA水平明显高于非耐药胱癌细胞株T24组,差异有统计学意义(P <0. 05);膀胱癌组UCA1、miR-141 mRNA明显高于癌旁正常组,差异有统计学意义(P <0. 05); UCA1、miR-141 mRNA水平与肿瘤分级、肌层浸润、肿瘤分期、发病特点呈正相关(r值分别为0. 53、0. 57、0. 61、0. 41、0. 58、0. 43、0. 56、0. 40,P <0. 05);UCA1-组、miR-141-组、UCA1-/miR-141-组3年生存率及生存期均明显高于UCA1+组、miR-141+组、UCA1+/miR-141+组,差异均有统计学意义(P <0. 05)。结论 UCA1、miR-141在膀胱癌组织的表达量高于正常膀胱组织,UCA1、miR-141在膀胱癌的发生发展过程中起促进作用; UCA1、miR-141低表达患者能获得较好的预后。Objective To investigate the expression of UCA1 and miR141 in bladder cancer cells and bladder cancer tissues and its correlation with prognosis of patients.Methods The expression levels of UCA1 and miR141 mRNA in human bladder epithelial immortalized cell line SVHUC1 and human bladder cancer cell lines including BIU87,EJ,T24,T24MMC,T24ADM were detected,respectively.Meanwhile the expression levels of UCA1 and miR141 mRNA in bladder cancer tissues and tumor-adjacent normal tissues in 78 patients with bladder cancer were measuredand the correlation between the expression levels and prognosis of patients was analyzed.Results The expression levels of UCA1 and miR141 mRNA in bladder cancer cell lines BIU87,EJ,T24,T24MMC,T24ADM groups were significantly higher than those in bladder epithelial immortalized cell lines SVHUC1 group(P<0.05).The expression levels of UCA1 and miR141 mRNA in drug-resistant human bladder cancer cell line T24MMC,T24ADM groups were significantly higher than those in non-resistant bladder cancer cell line T24 group(P<0.05).Moreover,the expression levels of UCA1 and miR141 mRNA in bladder cancer group were significantly higher than those in adjacent normal tissues group(P<0.05).Moreover the expression levels of UCA1 and miR141 mRNA were positively correlated with tumor grade,myometrial invasion,tumor staging and pathogenesis(r=0.53,0.57,0.61,0.41,0.58,0.43,0.56,0.40,respectively P<0.05).In addition the 3-year survival rate and survival duration in UCA1 group,miR141group,UCA1/miR141group were significantly higher than those in UCA1+group,miR141+group,UCA1+/miR141+group(P<0.05).Conclusion The expression levels of UCA1 and miR141 in bladder cancer tissues are higher than those in normal bladder tissues.The UCA1 and miR141 play an important role in the pathogenesis and development of bladder cancer.The patients with lower expression of UCA1 and miR141 have better prognosis.
