摘 要:【目的】研究铝箔袋真空密封和尼龙网袋非密封方法包装贮藏的临沧铁壳麦种子生活力指标、贮藏期、a-淀粉酶和脂肪氧化酶(LOX)活性及其基因相对表达量间的关系,了解其a-淀粉酶和LOX活性及其基因相对表达量变化影响种子贮藏期的酶学机制,为更好地保存临沧铁壳麦等云南小麦亚种资源提供参考依据。【方法】将临沧铁壳麦种子真空密封于铝箔袋和非密封包装于尼龙网袋内,在昆明室内温、湿度条件下贮藏63、125、189、210、252和365 d。采用标准发芽试验、3,5-二硝基水杨酸、分光光度计和实时PCR(RT-PCR)方法,分别测量种子的生活力指标、a-淀粉酶和LOX活性及其基因相对表达量,并进行简单相关和逐步回归分析。【结果】真空密封包装的种子在贮藏125 d后,发芽势为78.3%,发芽率为96.7%,加权发芽指数为0.622,发芽指数为30.933,活力指数为539.1,而非密封包装的种子在贮藏125 d后发芽势、加权发芽指数及发芽指数已明显下降,至189 d时观测的5项生活力指标均出现断崖式下降,分别只有125 d时的14.9%~25.0%。两种方法包装贮藏后的种子,a-淀粉酶和LOX活性无明显差异。种子贮藏期与生活力指标、酶活性及其基因相对表达量的简单相关及逐步回归分析结果表明,在昆明室内温、湿度条件下,非密封方法包装贮藏的临沧铁壳麦种子贮藏期随加权发芽指数和LOX活性的增加而缩短,随a-淀粉酶活性和TaLOX3-4A基因相对表达量的增加而延长,但真空密封方法包装贮藏的种子,其贮藏期只随a-淀粉酶基因amy3相对表达量的减少而延长。【结论】真空密封在铝箔袋中的临沧铁壳麦种子具有更长的、保持高生活力的贮藏期,其贮藏期不随a-淀粉酶和LOX活性变化而发生变化。【Objective】Relationship between vacuum sealed package by aluminium foil bag and non-sealed package by nylon mesh bag in viability index,periods of seed stored(PSS),activities of a-amylase and lipoxygenase(LOX)and gene relative expression of the Lincang hulled wheat seeds stored in the two packages was studied.Enzymatic mechanism of activities of a-amylase and LOX and gene relative expression affecting PSS was analyzed to provide reference to better store Triticum aestivum ssp.Yunnanense including Lincang hulled wheat.【Method】Lincang hulled wheat seeds were put into vacuum sealed aluminum foil bag and into non-sealed nylon mesh bags,stored at indoor temperature and relative humidity conditions in Kunming for 63,125,189,210,252 and 365 d.Then standardized germination test,3,5-dinitrosalicylic acid(3,5-DNSA),spectrophotometer and real time PCR(RT-PCR)were used to measure respectively seeds viability index,activities of a-amylase and LOX,and relative expression level(REL)of the seeds.The simple correlation analysis and stepwise regression were also conducted.【Result】The results showed that the seeds packaged in vacuum sealed method still had very high viability indexes after storing 125 d,whose germination force(GF),germination percent(GP),weighted germination index(WGI),germination index(GI)and vigor index(VI)were 78.3%,96.7%,0.622,30.933,and 539.1 respectively.But for the seeds after storing 125 d with non-sealed package,GF,WGI and GI had very obvious decline,all the five viability indexes occurred cliff collapse when seeds were stored up to 189 d,only occupied by 14.9%-25.0%of those stored for 125 d.No obvious difference was found in the activities of a-amylase and LOX between seeds packaged by the two methods.The simple correlation and stepwise regression analysis between PSS and seeds viability index,activities of a-amylase and LOX,and REL of genes indicated that,under indoor temperature and relative humidity conditions in Kunming,PSS with non-sealed package way were shortened with increase of WGI an
