摘  要:目的研究全反式维甲酸(ATRA)联合第三代铂类抗癌化合物对人肝癌SMMC-7721细胞的抑制作用。方法肝癌SMMC-7721细胞复苏及培养,实验共分为对照组、ATRA组、L-OHP组、联合组4组,方法为对照组不加药将等体积的培养液加入,ATRA组将10-5mol/L ATRA加入,L-OHP组将20 mg/L奥沙利铂(L-OHP)加入,联合组将10-5mol/L ATRA+20 mg/L L-OHP加入,MTT法测定细胞生长抑制率,流式细胞术测定细胞周期及凋亡情况,然后对4组不同时间点的A490值,G0/G1期、S期、G2/M期、细胞凋亡率进行统计分析。结果联合组和L-OHP组1、2、3 d随着用药剂量的提升A490值均逐渐降低(P <0. 05),均显著低于ATRA组和对照组(P <0. 05);联合组的G0/G1、G2/M均显著低于L-OHP组、ATRA组、对照组(P <0. 05),S期、细胞凋亡率均显著高于L-OHP组、ATRA组、对照组(P <0. 05)。结论 ATRA能够对人肝癌SMMC-7721细胞的生长进行有效抑制并对细胞凋亡进行诱导,联合第三代铂类抗癌化合物奥沙利铂能够促进奥沙利铂用量的减少,发挥协同作用。Objective To study the inhibitory effect of ATRA combined with third generation platinum anticancer compound on human hepatocellular carcinoma SMMC-7721 cells.Methods These cells were given with recovery and cultivation,they were randomly divided into control group,ATRA group,L-OHP group and combination group four groups,cells in control group did not add any drug,the same volume of culture medium was added,in ATRA group added 10-5 mol/L ATRA,the L-OHP group added 20 mg/L L-OHP,in combined group added 10-5 mol/L ATRA+20 mg/L L-OHP.The inhibition rate of cell growth had been detected by MTT assay,the cell cycle and apoptosis were determined by flow cytometry,and then the A490 at different time points,stage G0/G1,stage S,stage G2/M,apoptosis rates of these four groups were statistically analyzed.Results In 1 st d,2 nd d and 3 rd d,A490 value of combined group and L-OHP group had been decreased with the increased dosage(P<0.05),they were significantly lower than those of ATRA group and control group(P<0.05).Stages G0/G1 and G2/M of combined group were significantly lower(P<0.05).The S,apoptosis rate was significantly higher than those of L-OHP group,ATRA group and control group(P<0.05).Conclusion ATRA can effectively inhibit the growth of human hepatoma SMMC-7721 cells and induce apoptosis.Combination with third generation of platinum anticancer compound Oxaliplatin can reduce the dosage of oxaliplatin,thus it will play.synergitic action.
