摘　　要：目的研究熟地黄Rehmannia Radix Praeparata的化学成分及其体外生物活性。方法熟地黄水提取物采用正相硅胶、Sephadex LH-20、反相HPLC柱进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。以MTT法测定其肝细胞损伤保护活性。结果从中分离得到7个化合物,分别鉴定为isobonein (1)、地黄素A (2)、苁蓉素(3)、地黄素D (4)、地黄呋喃(5)、异毛蕊花糖苷(6)、肉苁蓉苷F (7)。化合物1、2、4、7可提高HL-720细胞的成活率。结论化合物1、3、7为首次从该属植物中分离得到,化合物1、2、4、7表现出较明显的体外保肝活性。AIM To study the chemical constituents of Rehmannia Radix Praeparata and biological activities in vitro. METHODS The aqueous extract from Rehmannia Radix Praeparata was isolated and purified by normal-phase silica column,Sephadex LH-20 and reversed-phase HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data. Their hepatocyte protection activities were determined by MTT. RESULTS Seven compounds were isolated and identified as isobonein(1),rehmaglutin A(2),cistanin(3),rehmaglutin D(4),jiofuran(5),isoacteoside(6) and cistanoside F(7). Compounds1,2,4 and 7 could improve the survival rate of HL-720 cells. CONCLUSION Compounds 1,3 and 7 are isolated from genus Rehmannia for the first time,compounds 1,2,4 and 7 have obviously hepatocyte protection bioactivities in vitro.
