摘 要:目的观察不同浓度黄芪注射液对高糖环境下肾小管上皮细胞(HK-2)凋亡和核转录因子κB (NF-κB)蛋白表达的影响,探讨黄芪注射液浓度与细胞凋亡率、NF-κB蛋白表达间的关系。方法肾小管上皮细胞用无血清培养基同步化24 h后随机分为低糖组、高糖组、高糖+2μg/m L黄芪注射液组、高糖+20μg/m L黄芪注射液组、高糖+200μg/m L黄芪注射液组,每组设3个复孔。细胞培养48 h和72 h后收集细胞及细胞上清液,用流式细胞仪测定各组细胞凋亡率,Western blot方法测定各组NF-κB蛋白表达量,分析黄芪注射液浓度、细胞凋亡率、NF-κB蛋白表达之间的相关性。结果细胞培养48 h和72 h,低糖组和黄芪注射液各干预组细胞凋亡率及NF-κB蛋白表达量均明显低于高糖组(P均<0. 05)。黄芪注射液各干预组间细胞凋亡率及NF-κB蛋白表达量比较差异均有统计学意义(P均<0. 05),高糖+200μg/m L黄芪注射液组最低。黄芪注射液浓度与细胞凋亡率、NF-κB蛋白表达量呈负相关,NF-κB蛋白表达量与细胞凋亡率呈正相关。结论黄芪注射液能抑制高糖诱导的肾小管上皮细胞凋亡和NF-κB蛋白表达,黄芪注射液浓度、细胞凋亡率、NF-κB蛋白表达之间有显著相关性。Objective It is to observe the influence of different dose of Astragalus on expression of nuclear factor-κB(NF-κB)and cell apoptosis induced by high glucose in human proximal renal tubular epithelial cells(HK-2),and to explore the relationship between the concentration of Astragalus injection and the apoptosis rate and NF-κB protein expression.Methods Renal tubular epithelial cells were synchronized with serum-free medium for 24 hours and then randomly divided into low glucose group,high glucose group,high glucose+2μg/mL Astragalus injection group,high glucose+20μg/mL Astragalus injection group,high glucose+200μg/mL Astragalus injection group,each group had 3 duplicate holes.The cells and cell supernatant were collected after 48 h and 72 h of cell culture.The apoptosis rate of each group was determined by flow cytometry.The expression of NF-κB protein was determined by Western blot.The relationship among concentration of Astragalus injection,apoptosis rate and NF-κB protein expression was analyzed.Results When the cells were cultured for 48 h and 72 h,the apoptosis rate and the expression of NF-κB of low glucose group and every Astragalus injection treatment group were significantly lower than that of high glucose group(P<0.05).There was significant difference in apoptosis rate and the expression of NF-κB among Astragalus injection groups(P<0.05),those were the lowest in high glucose+200μg/mL Astragalus injection group.The concentration of Astragalus was negatively correlated with the apoptosis rate of cell and expression of NF-κB,the expression of NF-κB was positively correlated with apoptosis rate.Conclusion Astragalus injection can inhibit the apoptosis of renal tubular epithelial cells induced by high glucose and the expression of NF-κB protein.There is a significant correlation between the concentration of Astragalus injection,apoptosis rate and NF-κB protein expression.
