摘 要:目的检测miR-93在食管鳞癌组织和细胞中的表达情况,并分析其与患者临床病理特征及预后的关系。方法采用实时荧光定量PCR(qRT-PCR)法检测miR-93在食管鳞癌细胞系和正常食管细胞系中的表达情况,并且收集110例份食管鳞癌组织及癌旁组织标本,采用qRT-PCR法检测组织中miR-93的表达情况,分析miR-93表达与患者临床病理特征及预后的关系。结果 qRT-PCR结果显示,人食管鳞癌细胞系KYSE410、ECA-109、KYSE-510中miR-93的表达水平较人食管正常细胞系Het-1A升高(P均<0.01),食管鳞癌组织中miR-93的表达水平较癌旁组织升高(P<0.01),食管鳞癌组织中miR-93表达与患者的肿瘤分化程度、淋巴结转移及临床分期密切相关(P均<0.05),而与性别、年龄、肿瘤直径及肿瘤位置无关(P均>0.05),Kaplan-Meier生存分析显示,miR-93高表达患者的预后较差(P<0.01),Cox回归分析结果表明,淋巴结转移、临床分期、miR-93表达是影响患者预后的独立危险因素(P均<0.05)。结论 miR-93在食管鳞癌组织和细胞中表达升高,其异常高表达与食管鳞癌的发生、发展相关,是影响患者预后的独立危险因素。Objective To detect the expression of miR-93 in esophageal squamous cell carcinoma(ESCC) tissues and cells,and to analyze its relationship with clinicopathological features and prognosis.Methods Quantitative real-time fluorescence PCR(qRT-PCR) was used to detect the expression of miR-93 in esophageal squamous cell carcinoma cells and normal esophageal cells.Totally 110 specimens of esophageal squamous cell carcinoma and adjacent tissues were collected.The expression of miR-93 in the tissues was detected by qRT-PCR.The relationship between the expression of miR-93 and clinical pathological characteristics and prognosis of patients was analyzed.Results The results of qRT-PCR showed that the expression level of miR-93 in the human esophageal squamous cell carcinoma cell lines KYSE410,ECA-109 and KYSE-510 was higher than that in the human esophageal normal cell line Het-1 A(P<0.01).The expression of miR-93 in the esophageal squamous cell carcinoma was higher than that in the adjacent tissues(P<0.01).The expression of miR-93 in the esophageal squamous cell carcinoma was closely related to the degree of differentiation,lymph node metastasis,and clinical stage(all P<0.05),but was not related to sex,age,tumor diameter or location(P>0.05).KaplanMeier survival analysis showed that the prognosis of patients with high miR-93 expression was poor(P<0.01).Cox regression analysis showed that lymph node metastasis,clinical stage,and expression of miR-93 were independent risk factors for prognosis(all P<0.05).Conclusion The abnormal high expression of miR-93 is associated with the occurrence and development of esophageal squamous cell carcinoma,and is an independent risk factor affecting the prognosis of patients.
