摘　　要：目的验证miR-98可抑制肺癌患者B10细胞IL-10的表达。方法收集肺癌组和对照组外周血各10例,分离培养CD19B淋巴细胞,流式细胞检测B10细胞数,RT-PCR检测B10细胞miR-98和IL-10mRNA的表达,制备miR-98脂质体及建立肺癌小鼠模型,观察miR-98脂质体对小鼠瘤体生长的抑制作用。结果肺癌组外周血B10细胞miR-98显著低于对照组,IL-10 m RNA水平显著高于对照组,B10细胞miR-98与IL-10 mRNA间存在负相关。结论在miR-98高表达的环境下,B淋巴细胞IL-10表达降低,对荷瘤小鼠应用脂质体包埋miR-98能减慢肺癌生长。Objective This study tested a hypothesis that miR-98 suppresses expression of IL-10 in B cells of subjects with lung cancer.Methods We collected blood samples from patients with lung cancer and cultured the isolated CD19 B cells.Frequency of peripheral B10 cells was measured,as well as miR-98 and IL-10 mRNA in B cells.The miR-98 carrying liposomes and a lung cancer.bearing mouse model were developed to tested effects of miR-98.carrying liposomes on suppression of IL-10 in B cells.Results miR-98 levels were significantly less in lung cancer patients,whereas IL-10 mRNA levels were significantly higher as compared with healthy controls.Conclusion Up.regulation of miR-98 inhibits expression of IL-10 in B cells,which may contribute to inhibit the lung cancer tolerance in the body.
